乳腺癌患者血浆中盐酸吉西他滨的血药浓度测定及其药代动力学研究

摘要 目的本研究旨在建立一种测定血浆中盐酸吉西他滨含量的方法,并探讨其在乳腺癌患者中的药代动力学特征及相关参数,以期为临床医生制定个体化给药方案提供科学依据。方法采用HPLC法测定盐酸吉西他滨的血药浓度,以此为基础使用DAS软件计算盐酸吉西他滨在乳腺癌患者中的药代动力学参数,并对其进行房室模型拟合。结果盐酸吉西他滨的保留时间为17.5 min左右,线性范围为0.04~10.0μg·mL^(-1);通过药代动力学分析发现,盐酸吉西他滨在乳腺癌患者体内的代谢过程可用二室模型进行拟合。结论本实验操作简便,结果准确,准确度与精密度均符合方法学要求,可用作人体血浆中盐酸吉西他滨的浓度测定方法及药动学研究。在乳腺癌患者血浆中的盐酸吉西他滨药代动力学符合二室模型拟合。 OBJECTIVE The aim of this study was to establish a method for the determination of gemcitabine hydrochloride in plasma,and to investigate its pharmacokinetic characteristics and related parameters in breast cancer patients,in order to provide scientific basis for clinicians to make individualized drug administration regi-men.METHODS The plasma concentration of gemcitabine hydrochloride was determined by HPLC.On this basis,DAS software was used to calculate the pharmacokinetic parameters of gemcitabine hydrochloride in breast cancer patients,and the atrioventricular model was fitted.RESULTS The retention time of gemcitabine hydrochloride was about 17.5 min.The linear range of gemcitabine hydrochloride for this method is 0.04-10.0μg·mL^(-1).Through pharmacokinetic analysis,it was found that the metabolic process of gemcitabine hydrochloride in breast cancer pa-tients could be fitted with a two-compartment model.CONCLUSION The experiment is simple and accurate.The accuracy and precision meet the methodological requirements.It can be used for the determination of gemcitabine hydrochloride concentration in human plasma and pharmacokinetic study.The pharmacokinetics of gemcitabine hydrochloride in the plasma of breast cancer patients fit the two-compartment model.
