摘要
目的评估新型复合水溶性膳食纤维(novel composite soluble dietary fiber,NCSDF)对db/db小鼠的降糖效果,并探讨其是否通过肠道菌群-短链脂肪酸(short-chain fatty acids,SCFAs)-胰高血糖素样肽-1(glucagon-like peptide-1,GLP-1)信号轴发挥降糖作用。方法将8周龄雄性db/db小鼠分为模型组、低、中、高剂量NCSDF给药组,以及db/m小鼠对照组,每组10只。连续给药8周后,检测各组小鼠的空腹血糖、胰岛素、GLP-1水平,并通过16S rRNA测序和SCFAs检测分析肠道菌群结构和代谢物水平。结果NCSDF干预可降低db/db小鼠的空腹血糖水平,改善胰岛素抵抗,并上调血清GLP-1水平。此外,NCSDF干预还可调节肠道菌群结构,增加拟杆菌(Bacteroides)、毛螺菌(Lachnospiraceae)和普雷沃菌(Prevotellaceae)等产SCFAs菌属的丰度,并提高肠道内容物中乙酸、丙酸、丁酸、异丁酸和戊酸的浓度。结论NCSDF可能通过调节肠道菌群-SCFAs-GLP-1信号轴,促进GLP-1的释放,增强外周组织胰岛素敏感性,从而发挥降糖作用。
Objective To evaluate the hypoglycemic effect of novel composite soluble dietary fiber(NCSDF)on db/db mice,and explore whether it exerts its hypoglycemic action through the gut microbiota-short-chain fatty acids(SCFAs)-glucagon-like peptide-1(GLP-1)signaling axis.Methods The 8 weeks-old male db/db mice were divided into a model group,low-,medium-and high-dose NCSDF treatment groups,and a control group of db/m mice,with 10 mice in each group.After continuous administration for 8 weeks,fasting blood glucose,insulin and GLP-1 levels were measured in each group.The gut microbiota structure and metabolite levels were analyzed using 16S rRNA sequencing and SCFAs detection.Results NCSDF intervention reduced fasting blood glucose levels,improved insulin resistance,and upregulated serum GLP-1 levels in db/db mice.Additionally,NCSDF intervention modulated the gut microbiota structure,increasing the abundance of SCFA-producing bacterial genera such as Bacteroides,Lachnospiraceae and Prevotellaceae,and elevated the concentrations of acetic acid,propionic acid,butyric acid,isobutyric acid and valeric acid in the intestinal content.Conclusion NCSDF may promote GLP-1 release and enhance insulin sensitivity in peripheral tissues by modulating the gut microbiota-SCFAs-GLP-1 signaling axis,thereby exerting its hypoglycemic effect.
