摘要
分蘖数是影响高粱株型和产量的关键因素,定位影响该性状的主效数量性状基因座(QTL)可为高粱的分子遗传改良提供依据。本研究以美国高粱品种BTx623与贵州酒用高粱品种红缨子杂交构建的包含205个家系的重组自交系(RIL)群体为材料,在2020年和2021年贵阳(2020GY、2021GY)、2020年安顺(2020AS)3个环境条件下调查分蘖数,并利用构建的高密度遗传图谱,采用完备区间作图法(ICIM)开展QTL定位。结果表明,在第1、2、5号染色体上共检测到5个与高粱分蘖数密切相关的QTL,涉及5个染色体区段。其中有2个主效QTL在2个环境中被重复检测到,分别为qTN3.2和qTN5.1,前者与已知的分蘖数基因OsASI1一致,而后者所在区间与水稻DHT1基因同源。同时分析了两个候选基因在高粱不同生育时期各组织中的表达模式以及在242份高粱种质中的单倍型变异情况,结果表明两个候选基因的表达呈现不同的时空特性,其序列变异与分蘖数表型显著相关。本研究结果可为后续高粱分蘖数基因的克隆和功能验证提供理论依据。
Tiller number(TN)is a key factor affecting plant architecture and yield in sorghum.Mapping major-effect quantitative trait loci(QTLs)controlling this trait can provide a theoretical basis for molecular genetic improvement of sorghum.In this study,a recombinant inbred line(RIL)population comprising 205 families derived from the cross between the American sorghum cultivar BTx623 and the Guizhou liquor-making sorghum cultivar Hongyingzi was used as experimental material.TN phenotypes were investigated under three environmental conditions,including Guiyang in 2020 and 2021(2020GY,2021GY)and Anshun in 2020(2020AS).Based on a constructed high-density genetic linkage map,QTL mapping was performed using the inclusive composite interval mapping(ICIM)method.The results showed that a total of five TN-associated QTLs were detected on chromosomes 1,2 and 5,corresponding to five chromosomal segments.Among them,two major-effect QTLs,qTN3.2 and qTN5.1,were repeatedly identified across two environments.qTN3.2 was consistent with the known tiller number gene OsASI1,while the genomic interval harboring qTN5.1 contained a homolog of the DHT1 gene.Meanwhile,the expression patterns of these two candidate genes in various tissues at different growth stages of sorghum and the haplotype variations in 242 sorghum germplasm accessions were analyzed.The results of this study could provide a theoretical foundation for subsequent cloning and functional verification of tiller number-related genes in sorghum.
