二倍体和三倍体陆封型马苏大麻哈鱼肌肉的蛋白质组学比较

摘要 为解析二倍体与三倍体陆封型马苏大麻哈鱼肌肉蛋白质组学差异,本实验选取相同养殖条件下的2龄二倍体(700~800 g)和三倍体(1250~1500 g)各9尾,取背鳍以下侧线以上肌肉进行基于DIA模式的蛋白质组学分析,结果共筛选出492个差异表达蛋白(245上调、247下调),其中检测到4个与肌肉功能相关的关键蛋白。结果发现,三倍体组肌球蛋白重链9(myosin-9,MYH9)、肌球蛋白轻链3(myosin light polypeptide 3,MYL3)、肌红蛋白(myoglobin,MB)和胶原蛋白ⅩⅩⅤⅢ型(collagen,typeⅩⅩⅤⅢ,COL28)的蛋白丰度显著高于二倍体组。将差异表达蛋白进行GO富集分析,显著富集的条目共57个,其中生物过程34个,主要涉及对硒离子的响应、对过氧化氢的响应和对活性氧的响应等功能条目。细胞组成10个,主要涉及呼吸链复合体Ⅲ、线粒体呼吸链复合体Ⅲ和蛋白酶体核心复合物等功能条目。分子功能13个,主要涉及谷胱甘肽二硫还原酶(NADP)活性、谷胱甘肽二硫化物氧化还原酶活性和氧化还原酶活性等功能条目,上述主要涉及的条目共有谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、细胞色素b(cytochrome b,CYTB)、蛋白酶体(proteasome,PSM)和谷胱甘肽还原酶(glutathione reductase,GR)4个蛋白参与。KEGG显著富集通路共224条,主要富集在脂肪的消化和吸收、MAPK信号通路和肌醇磷酸盐代谢等通路上。上述主要富集到的通路中共享的差异表达蛋白包括载脂蛋白A-I(apolipoprotein A-I,APOA-1)、磷脂酸磷酸酶3(phospholipid phosphatase 3,PLPP3)、14-3-3ε(14-3-3 protein epsilon,YWHAE)、甘油-3-磷酸脱氢酶NAD+(glycerol-3-phosphate dehydrogenase,GPDH)和磷酸丙糖异构酶(triosephosphate isomerase,TIM)5个蛋白。推测三倍体肌肉可能通过影响抗氧化系统和脂代谢等过程发挥作用,这为三倍体技术对陆封型马苏大麻哈鱼肌肉生长及品质改良的分子机制提供了重要的科学依据,并为进一步的遗传育种研究提供了关键数据。 In order to analyze proteomic differences between diploid and triploid Oncorhynchus masou masou,this experiment selected nine 2-year-old diploid fishs(700-800 g)and nine triploid fishs(1,250-1,500 g)under the same aquaculture conditions.Muscle samples were taken from below dorsal fin and above lateral line for proteomic analysis based on DIA(Data-Independent Acquisition)model.Results identified a total of 492 differential proteins(245 upregulated and 247 downregulated).In differential expressed proteins,four key proteins related to muscle function were detected.Results showed that the protein abundance of key muscle-related proteins such as myosin heavy chain 9(MYH9),myosin light polypeptide 3(MYL3),myoglobin(MB)and collagen typeⅩⅩⅤⅢ(COL28)were significantly higher in triploid group compared to diploid group.A total of 57 significantly enriched terms were identified through GO enrichment analysis of differentially expressed proteins.There were 34 biological processes,primarily involving terms related to response to selenium ion,response to hydrogen peroxide and response to reactive oxygen species.There were 10 cellular processes,mainly involving respiratory chain complexⅢ,mitochondrial respiratory chain complex III and proteasome core complex.There were 13 molecular functions,mainly involving glutathione-disulfide reductase(NADP)activity,glutathione disulfide oxidoreductase activity and oxidoreductase activity.The aforementioned key terms involve four proteins:glutathione peroxidase(GSH-Px),cytochrome b(CYTB),proteasome(PSM),and glutathione reductase(GR).KEGG showed significant enrichment in 224 pathways,primarily in fat digestion and absorption,MAPK signaling pathway and inositol phosphate metabolism.The shared differentially expressed proteins in major enriched pathways include apolipoprotein A-I(APOA-1),phospholipid phosphatase 3(PLPP3),14-3-3 protein epsilon(YWHAE),glycerol-3-phosphate dehydrogenase(NAD+)(GPDH)and triosephosphate isomerase(TIM).It was speculated that triploid muscles may exert their effects by influencing antioxidant systems and lipid metabolism processes.This study provided important scientific evidence for molecular mechanisms by which triploid technology improved the muscle growth and quality of O.masou masou.It also offered key data for further genetic breeding research.
