失巢凋亡抵抗与肿瘤转移研究进展

摘要 肿瘤转移通常是导致患者死亡的主要原因,而失巢凋亡抵抗作为肿瘤细胞脱离原发灶后存活的关键机制,在转移过程中发挥重要作用。正常上皮细胞在脱离细胞外基质后会经历失巢凋亡,但恶性肿瘤细胞通过激活多种信号通路及调控关键分子来逃避失巢诱导的凋亡,进而获得更强的侵袭和转移能力。此外,肿瘤微环境、细胞因子、代谢重编程及癌细胞与基质相互作用等因素也在失巢凋亡抵抗过程中发挥关键作用。近年来,失巢凋亡抵抗机制的研究持续深入,为抑制肿瘤转移提供了潜在的治疗靶点,如抑制整合素信号、干预抗氧化应激以及细胞代谢重塑策略等。深入研究细胞失巢凋亡抵抗的机制,将有助于开发针对性治疗策略,抑制癌细胞转移并改善患者预后。本文系统综述了失巢凋亡抵抗在癌细胞转移中的作用机制与对肿瘤转移的影响,以期为未来研究和临床干预提供新的思路。 Tumor metastasis is often the leading cause of patient mortality,and anoikis resistance serves as a critical mechanism enabling tumor cells to survive after detaching from the primary site,playing a significant role in the metastatic process.Normal epithelial cells typically undergo anoikis after detachment from the extracellular matrix.However,malignant tumor cells evade anoikis by activating various signaling pathways and regulating key molecules,thereby acquiring enhanced invasive and metastatic capabilities.Additionally,factors such as the tumor microenvironment,cytokines,metabolic reprogramming and interaction between cancer cells and the extracellular matrix also play crucial roles in the process of anoikis resistance.In recent years,research in the mechanisms of anoikis resistance has deepened,providing potential therapeutic targets for inhibiting tumor metastasis,such as suppressing integrin signaling,intervening in oxidative stress responses and employing strategies for metabolic reprogramming.A thorough investigation of the mechanisms underlying anoikis resistance will aid in the development of targeted therapeutic strategies to inhibit cancer cell metastasis and improve patient prognosis.This article systematically reviews the mechanisms of anoikis resistance in cancer cell metastasis and its impact on tumor progression,aiming to provide new insights into future research and clinical interventions.
