髓源性抑制细胞在小鼠实验性牙周炎中的作用研究

摘要 目的初步研究髓源性抑制细胞(MDSCs)在小鼠实验性牙周炎中的作用研究。方法小鼠随机分为空白对照组(CON组)与牙周炎组(1W、2W、4W组),分别在1周、2周、4周取材,Micro-CT扫描观测牙槽骨吸收程度,流式细胞仪检测牙龈及颌下淋巴结中MDSCs及亚群的数量变化。建立小鼠牙周炎模型后给予MDSCs抑制剂(吉西他滨组为GEM组、全反式维甲酸组为ACI组),牙周炎对照组(PER组),正常对照组(NOR组),Micro-CT扫描测量釉牙骨质界至牙槽嵴顶的距离(CEJ-ABC),评价牙槽骨的吸收情况。构建髓系细胞敲除小鼠(Lyz2 KO)建立牙周炎模型,通过Micro-CT扫描及HE染色分析牙槽骨吸收情况、观察牙槽骨形态变化。结果Micro-CT检测显示造模4周时,小鼠上颌颊侧牙槽骨较空白对照组明显吸收。流式结果显示造模4周后,牙周炎小鼠颌下淋巴结中MDSCs比例高于空白对照组,而牙龈中MDSCs比例低于空白对照组。给予MDSCs抑制剂后,牙周炎小鼠牙槽骨骨吸收明显增多。与野生型小鼠相比,髓系细胞敲除小鼠牙周炎模型中牙槽骨吸收明显增多。结论随着牙周炎的进展,牙龈中总MDSC数量减少,引流淋巴结中总MDSCs及亚群数量均增多。清除MDSCs或广泛敲除包含MDSCs在内的髓系细胞,均会加重牙周炎导致的牙槽骨吸收。 Objective To investigate the role of myeloid-derived suppressor cells(MDSCs)in experimental periodontitis in mice.Methods Mice were randomly divided into control group and periodontitis group.The samples were collected at 1,2 and 4 weeks.Micro-CT scanning was used to measure the alveolar bone resorption.The Flow cytometry was used to measure the amount of MDSCs in the gingival and submandibular lymph nodes.After periodontitis model was established,mice were given MDSCs inhibitor(gemcitabine group GEM,all-trans-retinoic acid group ACI),periodontitis control group(PER group)and blank control group(NOR group).Micro-CT scan was used to measure the distance from the cemento-enamel junction to aheolar bone crest(CEJ-ABC)and evaluate the absorption of alveolar bone.The model of periodontitis was established in myeloid cell knockout mice(Lyz2 KO).The absorption of alveolar bone was analyzed by Micro-CT scanning and HE staining.Results Micro-CT showed that the alveolar bone of the buccal side of the maxilla was more absorbed than that of the blank control group at 4 weeks.Flow cytometry analysis showed that the MDSCs ratio in submandibular lymph nodes of periodontitis mice was higher than that of CON mice,but the MDSCs ratio in gingiva was lower than that of CON mice.After administration of MDSCs inhibitor,alveolar bone resorption was significantly increased in periodontitis mice.Compared with wild-type mice,alveolar bone resorption was significantly increased in the myeloid cells knockout mice with periodontitis.Conclusions With the development of periodontitis,the number of MDSCs in gingiva was decreased,and the number of MDSCs and its subpopulations in draining lymph nodes was increased.Clearance of MDSCs aggravates periodontitis-induced alveolar bone resorption,which is exacerbated by widespread knockout of myeloid cells including MDSCs.
