摘要
目的:评价二十五味珊瑚丸不同极性提取物体内、体外对血脑屏障的作用效果,阐明其作用于血脑屏障的相关机制。方法:采用血脑屏障体内外模型,考察二十五味珊瑚丸不同极性提取物的穿透作用;采用ELISA试剂盒法,考察其作用机制。结果:水蒸气蒸馏组分改善血脑屏障通透性的作用最强,且可显著下调Claudin-5、E-cadherin、ABCB1和ABCC1的表达。结论:二十五味珊瑚丸可能通过其有效成分抑制MAPK相关信号通路,进而调控Claudin-5、E-cadherin、ABCB1和ABCC1的表达,改善血脑屏障的紧密结构并减少药物外排作用,进而增强血脑屏障的通透性。
Objective To evaluate the effects of different polarity extracts of Ershiwu Flavors Shanhu Pills on the blood-brain barrier(BBB)both in vivo and in vitro,and elucidate the underlying mechanisms.Methods BBB penetration effects of the extracts were investigated using in vivo and in vitro BBB models,while ELISA kits were employed to explore the mechanisms.Results The steam distillation fraction exhibited the strongest enhancement of BBB permeability and significantly downregulated the expression of Claudin-5,E-cadherin,ABCB1,and ABCC1.Conclusion Ershiwu Flavors Shanhu Pills may enhance BBB permeability by inhibiting the MAPK-related signaling pathway through its active components,thereby regulating the expression of Claudin-5,E-cadherin,ABCB1,and ABCC1.This mechanism improves the structural integrity of the BBB and reduces drug efflux.
