摘要
以采集的徐州市(XZ)、淮安市(HA)、苏州市(SZ)及南通市(NT)四地甜瓜霜霉病菌为试材,采用田间病害识别,病原显微形态观察及ITS序列分析等方法构建了系统发育树,研究了病原菌分类,明确江苏省甜瓜霜霉病病原菌类型,以期为甜瓜抗霜霉病种质选育及抗病机理研究提供参考依据。结果表明:4个地区甜瓜霜霉病菌序列相似度高达99%,均属于Pseudoperonospora cubensis;此外,课题组以收集的204份甜瓜种质为试材,开展了苗期和成株期霜霉病抗病性鉴定。综上,204份甜瓜种质霜霉病抗性筛选出高抗霜霉病种质3份(554、20144和1234),抗霜霉病种质40份,中抗霜霉病种质113份。
Taking the downy mildew pathogen of melon collected from four places,namely Xuzhou(XZ),Huai′an(HA),Suzhou(SZ)and Nantong(NT),as the test materials,by using methods such as field disease identification,microscopic morphology observation of the pathogen and ITS sequence analysis,a phylogenetic tree was constructed to study the classification of the pathogen,and to clarify the types of downy mildew pathogen of melon in Jiangsu Province,in order to provide reference for breeding downy mildew-resistant melon germplasm and investigating disease resistance mechanisms.The results showed that the sequence similarity of the downy mildew pathogen of melon in the four regions was as high as 99%,and all belonged to Pseudoperonospora cubensis.In addition,the research group used 204 melon germplasms collected as test materials to conduct disease resistance identification of downy mildew at the seedling stage and the mature plant stage.The results showed that among the 204 melon germplasms,3 varieties were screened out as highly resistant to downy mildew(554,20144 and 1234),40 varieties were moderately resistant to downy mildew,and 113 varieties were resistant to downy mildew.
