Role of polyunsaturated fatty acids in sarcopenia:Insights from Mendelian randomization analysis

摘要 目的:采用孟德尔随机化(MR)方法来探究多不饱和脂肪酸(PUFAs)与肌肉减少症之间潜在的因果联系。方法:MR分析使用了PUFAs的GWAS汇总统计数据,包括ω-3 PUFAs、ω-6 PUFAs以及ω-6与ω-3的比值(ω-6∶3),以及与肌肉减少症相关的性状,如四肢瘦体质量(ALM)、握力下降和步行速度。采用了逆方差加权(IVW)方法和两种额外的MR方法来建立稳健的因果推断。使用Cochrane'Q检验和MR-Egger截距检验评估异质性和水平多效性。进行敏感性分析以增强结果的稳健性和可靠性。结果:IVW方法显示,较高ω-6水平与ALM减少之间存在显著的遗传关联(β=-0.06,P=6.474×10^(-6))。在ω-3 PUFAs、ω-6∶3和肌肉减少症之间未发现显著关联。没有证据表明异质性或水平多效性影响MR分析(所有P>0.05)。此外,敏感性分析证实了所得因果关系的可靠性。结论:ω-6 PUFAs水平升高与肌肉减少症发生风险增加之间存在潜在的因果联系。这些结果强调了减少ω-6 PUFAs摄入的重要性,特别是在遗传上易患肌肉减少症的个体中。 Objective:To examine potential causal relationships between polyunsaturated fatty acids(PUFAs)and sarcopenia through Mendelian randomization(MR)analysis.Methods:The MR analy-sis used GWAS summary statistics for PUFAs,including omega-3 PUFAs(omega-3),omega-6 PUFAs(omega-6),and the omega-6 to omega-3 PUFA ratio(omega-6∶3),as well as for sarcopenia-related traits,such as appendicular lean mass(ALM),reduced hand grip strength,and walking speed.The primary inverse-variance weighted(IVW)method and two additional MR approaches were utilized to establish robust causal inference.Heterogeneity and horizontal pleiotropy were as-sessed using Cochrane's Q and MR-Egger intercept tests.Sensitivity analyses were conducted to en-hance the robustness and reliability of the results.Results:The IVW approach demonstrated a signifi-cant genetic association between higher omega-6 levels and reduced ALM(β=-0.06,P=6.474×10^(-6)).No significant associations were found between omega-3 levels,omega-6∶3,and sarcopenia-re-lated traits.There was no evidence of heterogeneity or horizontal pleiotropy affecting the MR analysis(all P>0.05).Additionally,the sensitivity analyses confirmed the robustness of the estimated causal relationship.Conclusion:Our findings suggest a potential causal link between elevated levels of ome-ga-6 PUFAs and an increased risk of sarcopenia.These results highlight the importance of reducing the intake of omega-6 PUFAs,particularly among individuals with a genetic susceptibility to sarcope-nia.
