摘要
目的:初步探讨七草清肤方治疗湿疹的作用机制。方法:运用超高效液相色谱-质谱联用技术(UHPLC-MS)鉴定七草清肤方中化学成分,筛选相关靶点,构建成分-疾病、蛋白互作网络并进行通路富集分析;利用2,4-二硝基氯苯诱导湿疹小鼠模型,观察该方治疗湿疹效果,采用酶联免疫吸附测定法(ELISA)检测炎症因子水平;结合UHPLC-MS分析湿疹小鼠皮肤差异代谢物及相关通路。结果:共鉴定出142种化学成分,筛选出TNF等130个潜在作用靶点,主要富集于TNF等信号通路。该方可明显改善湿疹小鼠皮损症状,降低湿疹严重指数评分和搔抓频率,降低TNF-α、IL-1β、IL-4、IL-6、IL-10水平,升高IFN-γ水平,差异有统计学意义(P<0.05,P<0.01)。筛选出29种差异代谢物,涉及21条通路,其中嘌呤代谢可能为关键通路。结论:基于网络药理学与代谢组学技术,初步预测了七草清肤方中槲皮素等成分通过多靶点抑制促炎因子、调节Th1/Th2失衡及干预嘌呤代谢通路,调控免疫-代谢紊乱治疗湿疹,为该方院内制剂的开发与临床应用提供了科学依据。
OBJECTIVE To preliminarily explore the therapeutic mechanism of Qicao Qingfu Prescription in treating eczema.METHODS The chemical constituents of Qicao Qingfu Prescription were identified using ultra-high performance liquid chromatography coupled with mass spectrometry(UHPLC-MS).Disease-related and drug-related targets were screened in databases.Using bioinformatics tools,the component-disease and protein-protein interaction(PPI)networks were constructed,followed by pathway enrichment analysis.An eczema mouse model was induced using 2,4-dinitrochlorobenzene(DNCB)to explore the therapeutic efficacy of Qicao Qingfu Prescription in treating eczema.Enzyme-linked immunosorbent assay(ELISA)was employed to detect the levels of inflammatory cytokines.UHPLC-MS was further applied to analyze differentially expressed metabolites and signaling pathways in the skin tissue of eczema mice.RESULTS A total of 142 chemical constituents were identified,and 130 potential targets,including tumor necrosis factor(TNF),were screened.Enrichment analysis revealed that the TNF signaling and other signaling pathways were significantly enriched in identified targets.Treatment of Qicao Qingfu Prescription in eczema mice significantly alleviated skin lesion symptoms,decreased the Eczema Area and Severity Index(EASI)and scratching frequency,lowered contents of tumor necrosis factor-alpha(TNF-α),interleukin(IL)-1β,IL-4,IL-6,and IL-10,and increased interferon-gamma(IFN-γ)levels(P<0.05,P<0.01).A total of 29 differentially expressed metabolites and 21 signaling pathways were identified.Purine metabolism emerged as a potential key pathway.CONCLUSION Integrated network pharmacology and metabolomics revealed that active constituents(e.g.,quercetin)in Qicao Qingfu Prescription ameliorate eczema through multi-target mechanisms like inhibiting pro-inflammatory cytokines,regulating type 1 helper T(Th1)/Th2 imbalance,and modulating purine metabolism to correct immune-metabolic dysregulation.These findings provide a scientific basis for the further development and clinical application of this inhouse formulation.
