YAP信号通路在子宫内膜纤维化中的作用研究

摘要 目的探讨YAP信号通路在子宫内膜纤维化中的作用及机制。方法体外培养正常子宫内膜基质细胞,设置对照组、纤维化组(TGF-β组)及维替泊芬组(VP组),VP组处理24 h及分别用雌二醇、醋酸甲羟孕酮、环磷酸腺苷作用72 h以模拟蜕膜化状态。免疫荧光染色实验观察正常子宫内膜组织及宫腔粘连患者组织中YAP及纤维化标志物的表达情况;Western blot检测纤维化标志物及YAP信号通路相关蛋白表达情况;实时荧光定量聚合酶链反应(RT-qPCR)实验检测各组CYR61、CTGF、HOXA10、LIF的mRNA表达水平,以及进行蜕膜化处理后各组PRL的mRNA表达水平。结果免疫荧光染色实验显示,宫腔粘连患者组织中YAP及COL1A1表达明显升高(P<0.05)。Western blot及RT-qPCR结果显示,与对照组相比,TGF-β组的纤维化标志物、YAP、HOXA10、LIF表达水平显著升高,PRL的mRNA表达水平表达显著降低(P<0.05);与TGF-β组相比,VP组结果呈相反趋势(P<0.05)。结论抑制YAP信号通路可抑制子宫内膜基质细胞的纤维化,且不会阻碍子宫内膜基质细胞的蜕膜化进程。 Objective To investigate the role and mechanism of the YAP signaling pathway in endometrial fibrosis.Methods Normal human endometrial stromal cells(HESCs)were cultured in vitro,with control group,fibrosis(TGF-β1)group,and Verteporfin(VP)group established.The latter group was treated for 24 hours and then subjected to estradiol,medroxyprogesterone acetate,and cyclic adenosine monophosphate for 72 hours to simulate the decidual state.Immunofluorescence staining was used to examine the expression of YAP and fibrosis markers in normal endometrial tissue and in tissue from patients with intrauterine adhesions.Western blot analysis was performed to detect the expression of fibrosis markers and proteins associated with the YAP signaling pathway.Real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)experiment was used to measure mRNA expression levels of CYR61,CTGF,HOXA10,and LIF across all groups,as well as PRL mRNA expression levels following decidualization treatment.Results Immunofluorescence staining revealed significantly elevated expression of YAP and COL1A1 in tissue samples from patients with intrauterine adhesions(P<0.05).Westernblot and RT-qPCR results demonstrated that compared with the control group,the TGF-β group exhibited significantly elevated expression levels of fibrosis markers,YAP signaling pathway-related proteins,HOXA10,and LIF,along with significantly reduced mRNA expression of PRL(P<0.05).In contrast,the VP group showed opposite trends compared with the TGF-β group(P<0.05).Conclusion Inhibiting the YAP signaling pathway can regulate endometrial stromal cells,thereby exerting an anti-fibrotic effect without hindering the decidualization process of endometrial stromal cells.
