摘要
滩羊是宁夏“六特产业”之一,也是当地农牧民增收致富的重要支柱产业。本研究基于1×低深度重测序结合基因型填充的方法,对来自宁夏地区2个国家级核心育种场、1个国家级保种场和1个繁育场的794只滩羊进行了全基因组遗传多样性系统评估。结果表明,低深度重测序可实现较高的基因型填充质量(填充准确性达97.5%,基因型相关性为87.6%)。在此基础上,我们基于杂合度、LD衰减、ROH长度、亲缘关系与近交系数等指标评估了4个滩羊群体的遗传多样性。研究发现,两个核心育种场群体普遍具有较低的近交水平和较高的遗传多样性,反映其育种管理较为规范;保种场群体整体遗传多样性较高,反映其选种选配合理,保种措施得当,但存在少量高度近交个体,提示需关注潜在的近交积累风险;繁育场群体在多项指标上均表现出近交程度偏高、遗传多样性下降的趋势,应加强引种并优化选种选配策略。本研究为滩羊种质资源的保护利用及育种管理优化提供了重要的遗传学依据。
Tan sheep constitute one of Ningxia’s“six special industries”and serve as a vital pillar industry for local farmers and herders to increase income and achieve prosperity.This study conducted a comprehensive assessment of genomic diversity in 794 Tan sheep from two national core breeding farms,one national conservation farm,and one breeding farm in Ningxia.The analysis employed a 1×low-coverage resequencing approach combined with genotype imputation.Results indicate that low-coverage sequencing achieves high genotyping quality(97.5%imputation concordance and 87.6%genotype correlation).Based on this,we assessed genetic diversity across four Tan sheep populations using metrics including heterozygosity,LD decay,ROH length,kinship,and inbreeding coefficients.The study found that both core breeding farm populations exhibited generally low inbreeding levels and high genetic diversity,reflecting well-regulated breeding management.The conservation farm population exhibited overall high genetic diversity,indicating rational selection and mating practices along with effective conservation measures.However,the presence of a small number of highly inbred individuals suggests the need to monitor potential risks of inbreeding accumulation.The breeding farm population showed trends of elevated inbreeding and declining genetic diversity across multiple metrics,necessitating enhanced introduction of new bloodlines and optimization of selection and mating strategies.This study provides important genetic evidence for the conservation,utilization,and optimized breeding management of Tan sheep germplasm resources.
