脂质纳米药物递送系统

摘要 脂质纳米药物递送系统是20世纪90年代逐渐发展起来的一类新型药物递送平台,主要包括脂质体、脂质纳米粒、胶束、核酸-脂质复合物等。该系统在抗肿瘤、抗感染、疫苗和基因治疗等领域展现出重要的临床应用价值,尤其在核酸药物递送方面具有广阔的应用前景。本文系统阐述了脂质纳米药物递送系统的定义和分类,概述其上市及在研药物的发展现状,探讨了该系列技术的优势与挑战,并对未来研究方向进行了展望。总体而言,脂质纳米药物递送系统在药物递送领域具有极大的开发潜力。未来研究需致力于提升靶向递送效率、改善安全性、推进联合治疗策略等,以实现其在多种疾病治疗领域更精准、高效的应用。 Lipid-based nanodelivery drug systems,which emerged in the 1990s,constitute a novel class of drug delivery platforms,primarily including liposomes,lipid nanoparticles,micelles,and nucleic acid-lipid complexes.These systems have demonstrated significant clinical potential in diverse therapeutic areas,including oncology,infectious diseases,vaccine development,and gene therapy,with particularly promising prospects in nucleic acid drug delivery.This review systematically elaborates on the definition and classification of lipid-based nanodrug delivery systems,outlines the current developmental status of both marketed and investigational drugs,discusses their technological advantages and challenges,and presents an outlook on future research directions.Overall,lipid-based nanodelivery systems exhibit substantial potential in the field of drug delivery.Future research should focus on enhancing targeted delivery efficiency,improving safety profiles,and promoting combination therapy strategies to achieve more precise and effective applications in the treatment of various diseases.
