微粒化脱细胞真皮基质注射移植时适宜介质与混合比例的筛选 被引量:1

Screening of optimal medium and mixing ratio for micronized acellular dermal matrix injection transplantation

摘要 目的探索微粒化脱细胞真皮基质(mADM)注射移植时的适宜介质及混合比例。方法将0.5 g mADM与透明质酸钠凝胶(A1~A3组)、胶原蛋白(B1~B3组)、血清(C1~C3组)、PRP(D1~D3组)、颗粒脂肪(E1~E3组)、纳米脂肪(F1~F3组)分别按1∶1、1∶2、1∶4比例混合,设单纯介质组及18个混合组,检测不同混合物的注射压力。不同介质与mADM混合后均以1∶4比例组注射压力值最小,据此将mADM与不同介质的1∶4比例混合物分别对9只Wistar大鼠进行皮下注射,比较不同介质混合物的皮下注射压力,分析悬浮稳定性及mADM颗粒迁移速率。通过流变仪测定不同比例混合物的黏度,并利用仿真软件模拟针筒压缩过程,分析仿真注射压力与黏度变化的一致性。结果mADM与透明质酸钠凝胶、胶原蛋白、颗粒脂肪、纳米脂肪按不同比例混合后注射压力差异均有统计学意义(均P<0.05),且均以1∶4比例组的注射压力值最小(均P<0.05)。mADM与不同介质以1∶4比例混合后的大鼠皮下注射压力差异均有统计学意义(均P<0.05),注射压力从低到高的介质依次为PRP、血清、胶原蛋白、透明质酸钠凝胶、纳米脂肪、颗粒脂肪。在悬浮稳定性方面,mADM与不同介质以1∶4比例混合后,浊度稳定性指数(TSI)和颗粒迁移速率从低到高均依次为颗粒脂肪、纳米脂肪、PRP、透明质酸钠凝胶、血清、胶原蛋白。流变学指标检测结果显示,黏度从低到高依次为胶原蛋白、纳米脂肪、PRP、透明质酸钠凝胶、血清、颗粒脂肪,模拟推杆压力从低到高依次为PRP、血清、胶原蛋白、透明质酸钠凝胶、纳米脂肪、颗粒脂肪。结论胶原蛋白、纳米脂肪、PRP作为mADM的注射移植介质具有较大优势,mADM与不同介质的最佳混合比例为1∶4。 Objective To explore the appropriate medium and mixing ratio for injection transplantation of micronized acellular dermal matrix(mADM).Methodsm ADM(0.5 g)was mixed with sodium hyaluronate gel(Groups A1-A3),collagen(Groups B1-B3),serum(Groups C1-C3),platelet-rich plasma(PRP)(Groups D1-D3),granular fat(Groups E1-E3),and nano-fat(Groups F1-F3)at ratios of 1∶1,1∶2 and 1∶4,respectively.A pure medium group and 18 mixed groups were established,and the injection pressure of different mixtures was measured.Since the injection pressure of the 1∶4 ratio group was the lowest among all mixtures of different media and mADM,the 1∶4 ratio mixtures of mADM with different media were subcutaneously injected into 9 Wistar rats.The subcutaneous injection pressure of different medium mixtures was compared,and the suspension stability and migration rate of mADM particles were analyzed.The viscosity of mixtures with different ratios was determined using a rheometer,and the syringe compression process was simulated using simulation software.The consistency between simulated injection pressure and viscosity changes was analyzed.Results There were statistically significant differences in injection pressure among mixtures of mADM with sodium hyaluronate gel,collagen,granular fat,and nano-fat at different ratios(all P<0.05),and the injection pressure of the 1∶4 ratio group was the lowest in all cases(all P<0.05).There were statistically significant differences in subcutaneous injection pressure in rats among the 1∶4 ratio mixtures of mADM with different media(all P<0.05).The media ranked from lowest to highest injection pressure were PRP,serum,collagen,sodium hyaluronate gel,nano-fat,and granular fat.In terms of suspension stability,for the 1∶4 ratio mixtures of mADM with different media,the turbidity stability index(TSI)values and particle migration rates ranked from lowest to highest were granular fat,nano-fat,PRP,sodium hyaluronate gel,serum,and collagen.The results of rheological index detection showed that the viscosity ranked from lowest to highest was collagen,nano-fat,PRP,sodium hyaluronate gel,serum,and granular fat,and the simulated push rod pressure ranked from lowest to highest was PRP,serum,collagen,sodium hyaluronate gel,nano-fat,and granular fat.Conclusion Collagen,nano-fat,and PRP have significant advantages as injection transplantation media for mADM,and the optimal mixing ratio of mADM with different media is 1∶4.
