丹参“发汗”前后色度值与主要化学成分相关性研究

Correlation Between Chromatic Values and Active Components Content in Danshen(Salvia Miltiorrhiza)With or Without Sweating Treatments

摘要 目的探讨丹参“发汗”前后色度值与主要活性成分含量变化,并建立两者之间的线性关系,以期为丹参“发汗”科学内涵解读及产地加工提供科学参考。方法分别采用色差仪、高效液相色谱(HPLC)技术测定丹参“发汗”前后色度值及主要化学成分含量,并对二者的相关性进行分析,建立色度值与主要成分含量之间的线性关系。结果丹参“发汗”后,Δa*值和ΔL*绝对值整体呈下降趋势,Δb*值整体升高,木栓层、皮层与木部中酚酸类成分含量以下降为主,皮部的脂溶性成分含量整体升高。脂溶性成分二氢丹参酮、丹参酮Ⅰ、丹参酮ⅡA、隐丹参酮与丹参色度值Δa*、Δb*显著正相关,与ΔL*值显著负相关;水溶性成分丹参素钠、迷迭香酸与丹参色度值Δa*、Δb*显著负相关,与ΔL*值显著正相关,丹酚酸A与三个色度值均呈正相关。主要活性成分与色度值回归方程分别为:Y(二氢丹参酮)=-4.684+0.457X_(Δa*)+0.107X_(Δb*)+0.074X_(ΔL*),Y(丹参酮Ⅰ)=-29.018+3.544X_(Δa*)+0.486X_(Δb*)+0.530X_(ΔL*),Y_((丹参酮ⅡA))=-12.733+1.242X_(Δa*)+0.391X_(Δb*)+0.325X_(ΔL*),Y(隐丹参酮)=-10.213+1.222X_(Δa*)+0.228X_(Δb*)+0.243X_(ΔL*),Y_((丹参素钠))=1.601+0.044X_(Δa*)-0.022X_(Δb*)+0.014X_(ΔL*),Y_((迷迭香酸))=7.464+0.217X_(Δa*)-0.091X_(Δb*)+0.112X_(ΔL*),Y(丹酚酸A)=-0.157+0.003 X_(Δa*)+0.009X_(Δb*)+0.004X_(ΔL*)。结论通过量化丹参“发汗”前后外在颜色性状、内在活性成分之间的关系,建立了一种快速鉴别“发汗”丹参品质的方法。 Objective In order to the provide reference for the interpretation of scientific connotation of sweating treatments and quality evaluation of Danshen(Salvia Miltiorrhiza)and the color values and the contents of 13 components between Danshen(Salvia Miltiorrhiza)with or without sweating treatments were explored and the correlation was also established.Methods The color values were measured with the color difference meter and the contents of 13 main components were determined by HPLC.Their correlation and regression equations between components and the color value were analyzed.Results After sweating treatments,the absolute values ofΔa*and_(ΔL*)decreased while_(Δb*)increased as a whole.Phenolic acids in cork layer,cortex and wood parts mainly decreased,and the content of aliphatic acid in bark increased as a whole.The aliphatic acid components,including dihydrotanshinone,tanshinoneⅠ,tanshinoneⅡA and cryptotanshinone,were significantly positively correlated with the color values ofΔa*and_(Δb*)of Danshen(Salvia Miltiorrhiza),and negatively correlated with_(ΔL*).The water-soluble components Danshensu sodium and rosmarinic acid were negatively correlated with the color valuesΔa*and_(Δb*)of Danshen(Salvia Miltiorrhiza),and positively correlated with_(ΔL*),while salvianolic acid A was positively correlated with all three color values.The regression variances of active components and color values were Y(dihydrotanshinone)=-4.684+0.457X_(Δa*)+0.107X_(Δb*)+0.074X_(ΔL*),Y(tanshinoneⅠ)=-29.018+3.544X_(Δa*)+0.486X_(Δb*)+0.530X_(_(ΔL*)),Y(tanshinoneⅡA)=-12.733+1.242X_(Δa*)+0.391X_(Δb*)+0.325X_(ΔL*),Y(cryptotanshinone)=-10.213+1.222X_(Δa*)+0.228X_(Δb*)+0.243X_(ΔL*),Y(Danshensu sodium)=1.601+0.044X_(Δa*)-0.022X_(Δb*)+0.014X_(ΔL*),Y(rosmarinic acid)=7.464+0.217X_(Δa*)-0.091X_(_(Δb*))+0.112X_(ΔL*),Y(salvianolic acid A)=-0.157+0.003X_(Δa*)+0.009X_(Δb*)+0.004X_(ΔL*),respectively.Conclusion By quantifying the relationship between external color values and internal components contents of Danshen(Salvia Miltiorrhiza)with or without sweating treatments,a rapid method for identifying the quality of Chinese herbal medicine with sweating treatment was established.
