青梅果胶提取工艺及其抗氧化、降糖降脂活性

Extraction of green plum pectin and its antioxidant,hypoglycemic and hypolipidemic activities

摘要 为实现青梅的资源化利用,并开发可缓解果胶需求增长问题的新型果胶,本研究以青梅为原料,采用超声辅助柠檬酸提取青梅果胶的方法,探究不同提取条件对青梅果胶提取率的影响,并设计响应面实验优化提取工艺,对最优提取条件下提取的青梅果胶进行结构及理化性质分析;以商业果胶为参照,分析其抗氧化活性和降血糖降血脂能力。实验结果显示:柠檬酸浓度1.00 mol/L、超声功率150 W、料液比为1∶34(g/mL)、超声温度62℃、超声时间53 min的条件下,青梅果胶提取率最高,为26.92%,该条件下提取的果胶呈现块状结构,具有果胶特征吸收峰,分子量为219.94 kDa;青梅果胶对DPPH∙、ABTS^(+)∙、∙OH和O_(2)^(-)∙的清除能力以及对α-淀粉酶、胰脂肪酶的抑制能力均高于商业果胶。 In order to realize value-added utilization of green plum and develop new pectin fabrication process that can alleviate the increasing demand of pectin,the ultrasound-assisted citric acid extraction of green plum pectin was systematically optimized.Several different conditions were evaluated for increasing the rate of extraction of green plum pectin.Response surface experiments were designed to optimize the extraction process.The structure and physicochemical properties of the extracted green plum pectin under the optimal extraction conditions was analyzed and compared with commercial pectin.Its antioxidant activity,hypoglycemic and hypolipidemic ability were analyzed.The results showed that the maximum extraction rate of 26.92%was obtained under the conditions of citric acid concentration of 1.00 mol/L,ultrasonic power of 150 W,material-liquid ratio of 1:34 g/mL,ultrasonic temperature of 62℃,ultrasonic time of 53 min.The extracted pectin showed a block structure with pectin characteristic absorption peaks,and the molecular weight of 219.94 kDa.The scavenging capacity of green plum pectin against DPPH·,ABTS^(+)·,·OH and O_(2)^(−)∙,as well as its inhibitory ability onα-amylase and pancreatic lipase are all higher than those of commercial pectin.
