利用抗稻瘟病基因Pi57和Pi69改良软米品种云粳41的抗性

摘要 为满足生产上对水稻品种优质、抗病的需求,迫切需要在水稻育种中加强对优质与抗病性状的协同改良。以云粳41(YJ41)为轮回亲本,通过常规杂交、回交和自交的方法,结合分子标记辅助选择(MAS)技术,将抗稻瘟病基因Pi57和Pi69聚合到含有低直链淀粉含量调控基因du13(t)的优质粳稻品种YJ41中;同时,利用水稻基因芯片检测获得高回复率、携带单基因(Pi57或Pi69)和双基因(Pi57+Pi69)的BC_(2)F_(1)代导入系;此外,筛选出对YJ41感病的优势菌株,在温室条件下对单基因和双基因的BC_(2)F_(4)代纯合导入系及亲本材料进行人工接种,并开展抗病性鉴定评价。结果表明,接种对Pi57或Pi69基因材料表现致病的稻瘟病菌菌株后,携带双基因(Pi57+Pi69)的BC_(2)F_(4)代纯合单株均表现为抗病,而携带单基因(Pi57或Pi69)的BC_(2)F_(4)代纯合单株的抗感反应与供体亲本一致,说明抗性基因(Pi57+Pi69)聚合提高了YJ41的稻瘟病抗性水平,所获得的新种质对水稻优质抗病育种具有重要意义。 In order to meet the demand for high-quality and disease-resistant rice varieties in production,it is urgent to strengthen the synergistic improvement of quality and disease resistance in rice breeding.With Yunjing 41(YJ41)as the recurrent parent,the rice blast-resistant genes Pi57 and Pi69 were stacked into the high-quality japonica rice variety YJ41 carrying the low amylose content regulatory gene du13(t)through conventional hybridization,backcrossing,and self-crossing methods combined with molecular marker-assisted selection(MAS).Through rice gene chip detection,BC_(2)F_(1)introgression lines with high recovery rates and carrying single gene(Pi57 or Pi69)and double genes(Pi57 and Pi69)were obtained.The selected advantageous strains virulent to YJ41 were inoculated into the BC_(2)F_(4)homozygous introgression lines of both single gene and double genes and parental materials in the greenhouse,and the rice resistance to blast was evaluated.The results showed that after inoculation with rice blast strains that were pathogenic to the Pi57 or Pi69 gene materials,the BC_(2)F_(4)homozygous plants carrying the double genes(Pi57+Pi69)all demonstrated rice blast resistance.The BC_(2)F_(4)homozygous plants carrying single gene(Pi57 or Pi69)showed the same resistance/susceptibility as the donor parents.The results indicated that the pyramided Pi57+Pi69 genes improved the resistance of YJ41 to rice blast.These newly generated germplasm resources have important significance for high-quality and disease-resistant rice breeding.
