摘要
目的:通过星点设计-效应面优化法优化白芍簇晶显微特征常数的测定条件,以弥补国内常用均匀设计法的不足。方法:使用星点设计-效应面优化法进行实验设计优化,以白芍质量和甘油体积为考察因素,以白芍簇晶个数的RSD值为指标。求取二元线性拟合和二元非线性拟合,根据最佳模型绘制效应面,并选择最佳显微特征常数测定条件。结果:优化得到测定白芍簇晶显微特征常数的函数关系式为Y=42.12947-0.4481X_(1)-2.89185X_(2)+0.00282X_(1)^(2)+0.4821X_(2)^(2)-0.01604X_(1)X_(2)(R^(2)=0.601,P=0.000014)。最佳条件为白芍质量80 mg,甘油体积4.4 mL。结论:星点设计-效应面优化法具有精确度高以及实验次数少的优点,为中药显微特征常数的测定和优化提供了一种新的方法。
Objective:To optimize the determination conditions for the microscopic characteristic constants of crystalline clusters of Paeonia lactiflora by central composite design-response surface methodology(CCD-RSM),so as to address the limitations of the commonly used uniform design method in China.Methods:Central composite design-response surface methodology was adopted for experimental design optimization.The mass of Paeonia lactiflora and volume of glycerol were set as the factors under investigation,while the relative standard deviation(RSD)of the number of Paeonia lactiflora crystalline clusters served as the index.Binary linear fitting and binary nonlinear fitting were performed;the response surface was plotted based on the optimal model,and the best determination conditions for microscopic characteristic constants were selected.Results:The optimized functional relationship for determining the microscopic characteristic constants of Paeonia lactiflora crystalline clusters was obtained as follows:Y=42.12947-0.4481X_(1)-2.89185X_(2)+0.00282X_(1)^(2)+0.4821X_(2)^(2)-0.01604X_(1)X_(2)(R^(2)=0.601,P=0.000014).The optimal conditions were determined as follows:mass of Paeonia lactiflora=80 mg,volume of glycerol=4.4 mL.Conclusions:Central composite design-response surface methodology has the advantages of high accuracy and a small number of experiments,providing a new method for the determination and optimization of microscopic characteristic constants of traditional Chinese medicines.
