栀子水提物通过抑制多种炎症小体活化改善脂多糖诱导的急性肺损伤

The water extract of Gardenia jasminoides Ellisameliorates LPS-induced acute lung injury by inhibiting the activation of multiple inflammasomes

摘要 目的探讨栀子治疗急性肺损伤(ALI)的作用及机制。方法采用脂多糖(LPS)构建ALI小鼠模型,使用H&E染色评价肺组织病理变化,流式细胞计数法检测肺泡灌洗液中性粒细胞数量、ELISA及RT-qPCR检测血清和肺组织中炎症因子水平;联合BMDM构建多种炎症小体激活模型,通过免疫印迹法(WB)检测caspase-1 p20等蛋白表达和ASC寡聚化,酶联免疫吸附试验检测炎症因子IL-1β等含量,LDH试剂盒检测LDH释放量,流式细胞计数法检测mtROS变化。结果栀子可改善肺组织炎症浸润及细胞损伤,减少BALF中性粒细胞含量(P<0.001),减少血清和肺组织中IL-1β水平(P<0.001);还可显著抑制多种炎症小体激活;其主要是通过抑制ASC寡聚化和mtROS的产生而抑制炎症小体活化。结论栀子可通过减少mtROS释放及ASC寡聚抑制多种炎症小体激活,发挥治疗ALI的作用。 Objective To investigate the effect and mechanism of Gardenia jasminoides Ellis in treating acute lung injury(ALI).Methods An ALI mouse model was established using lipopolysaccharide.Pathological changes in lung tissue were assessed by H&E staining.Neutrophil count in bronchoalveolar lavage fluid(BALF)was detected via flow cytometry,while levels of inflammatory cytokines in serum and lung tissue were measured using ELISA and RT-qPCR.Bone marrow-derived macrophages were utilized to construct multiple inflammasome activation models.Western blotting was employed to detect protein expression of cysteinyl aspartate specific proteinase-1(caspase-1)p20 and apoptosis-associated speck-like protein containing a CARD(ASC)oligomerization.Interleukin-1 beta(IL-1β)levels were measured via ELISA kits,lactate dehydrogenase(LDH)release was assessed with LDH assay kit,and mitochondrial reactive oxygen species(mtROS)was quantified by flow cytometry.Results Gardenia jasminoides Ellis alleviated pulmonary inflammatory infiltration and cellular damage,significantly reduced neutrophil content in BALF(P<0.001),and decreased IL-1βlevels in serum and lung tissue(P<0.001).Gardenia jasminoides Ellis markedly suppressed activation of multiple inflammasomes,which inhibited inflammasome activation by suppressing ASC oligomerization and mtROS generation.Conclusion Gardenia jasminoides Ellis exerts a therapeutic effect on ALI by reducing the release of mtROS and the oligomerization of ASC,thereby inhibiting the activation of various inflammasomes.
