HOXC-AS2调控代谢重编程影响胰腺癌PANC-1细胞增殖、凋亡

HOXC-AS2 regulates metabolic reprogramming to affect the proliferation and apoptosis of pancreatic cancer PANC-1 cells

摘要 [目的]研究HOXC-AS2调控代谢重编程对胰腺癌细胞增殖及凋亡的影响。[方法]通过免疫组化实验对胰腺癌组织中HOXC-AS2的表达。将胰腺癌PANC-1细胞分为空白对照组、si NC组、si HOXC-AS2组。通过MTT实验和Brd U法分析胰腺癌PANC-1细胞增殖能力;流式细胞术分析胰腺癌PANC-1细胞的凋亡能力;蛋白免疫印迹实验分析PANC-1细胞中HOXC-AS2和糖酵解蛋白的表达,并分析PANC-1细胞中乳酸和腺嘌呤核苷三磷酸水平。[结果]HOXC-AS2在胰腺癌组织中高表达、癌旁组织中低表达(P<0.05)。与空白对照组及si NC组比较,si HOXC-AS2组的PANC-1细胞的HOXC-AS2表达降低,增殖能力减弱(P<0.05),凋亡率增加(P<0.05),糖酵解相关蛋白表达下调,乳酸和腺嘌呤核苷三磷酸水平降低(P<0.05)。[结论]HOXC-AS2在胰腺癌组织中表达上调。下调HOXC-AS2的表达能通过抑制胰腺癌的代谢重编过程进而抑制胰腺癌PANC-1细胞的增殖,促进PANC-1细胞凋亡。 [Objective]To investigate the effect of HOXC-AS2 on the proliferation and apoptosis of pancreatic cancer cells by regulating metabolic reprogramming.[Method]By immunohistochemical experiment on HOXC-AS2 expression in pancreatic cancer tissue.Pancreatic cancer cells PANC-1 can be divided into blank control group,si NC group,the si HOXC-AS2 group.Pancreatic cancer through the analysis of the determined by MTT experiment with the method of BrdU PANC-1 cell proliferation ability.By flow cytometry analysis ability to pancreatic cancer cell apoptosis PANC-1.The expression of HOXC-AS2 and glyeolytic proteins in PANC-1 cells was analyzed by Westerm blot experiments.and the levels of lactate and adeno-sine triphosphate in PANC-1 cells were analyzed.[Result]HOXC-AS2 is highly expressed in pancreatic cancer tisues,while its expression is low in pancreatic cancer adjacent tissues(P<0.05).Compared with the blank control group and si NC group,the expresion of HOXC-AS2 in PANC-1 cells in si HOXC-AS2 group was decreased,the proliferation ability was decreased(P<0.05),the apoptosis rate was increased(P<0.05),the expression of glycolysis-related proteins was down-regulated,and the levels of lactic acid and adenosine triphosphate were decreased(P<0.05).[Conclusion]HOXC-AS2 ex-pression is up-regulated in pancreatic cancer tissues.Down-regulation of HOXC-AS2 expression can inhibit the prolifera-tion of pancreatic cancer PANC-1 cells by suppressing the metabolic reprogramming process of pancreatic cancer cells and promote apoptosis of PANG-1 cells.
