BmNPV侵染过程中家蚕BmJun参与调控的靶基因鉴定

摘要 转录因子Jun是丝裂原活化蛋白激酶(mitogen-activated protein kinase,MAPK)信号通路下游重要的响应因子,可参与细胞生长、死亡和免疫反应等多种生命活动的调节。但对其在家蚕核型多角体病毒(Bombyx mori nucleopolyhedrovirus,Bm-NPV)侵染的家蚕细胞中的表达调控机制尚鲜见报道。为了深入探究BmJun在病毒侵染过程中对下游靶基因的调控作用,本研究采用ChIP-seq技术对家蚕BmN细胞中BmJun富集的DNA片段展开分析并进行鉴定。ChIP-seq结果发现大量的结合峰,除了坐落在基因间区外,还在基因启动子区有富集,并且在转录起始位点上下游1000 bp处的reads计数密度较大,暗示BmJun具有调控下游基因的作用。GO功能富集分析显示,BmJun结合的基因涉及组织器官发育、细胞质膜成分、信号受体活性、分子传感器活性、跨膜信号受体活性等功能。KEGG信号通路富集结果显示,BmJun结合相关基因在Wnt信号通路、昆虫激素合成、细胞色素P450对外源性药物的代谢作用、视黄醇新陈代谢、Hippo信号通路、黑色素生成、不饱和脂肪酸的生物合成、cAMP信号通路、mTOR信号通路、凋亡等通路有显著富集。最后,经过筛选获得并鉴定了BmJun在宿主基因组上结合的靶基因,根据已报道的Jun功能分析了BmJun的下游调控网络。研究结果将为BmNPV侵染期间家蚕转录因子BmJun参与调控相关功能的深入研究提供新参考。 Transcription factor Jun is an important downstream response factor of the mitogen-activated protein kinase(MAPK)signaling pathway,which can participate in the regulation of various life activities such as cell growth,death,and immune response.However,little is known about the regulatory mechanism of its expression in Bombyx mori nucleopolyhedrovirus(BmNPV)infected BmN cells.To explore the regulatory role of BmJun on downstream target genes during virus infection,in this study we analyzed and identified the DNA fragments enriched by BmJun in BmN cells through the ChIP-seq method.According to the ChIP-seq results,we found that a large number of binding peaks not only located in the intergenic regions but also enriched in the gene promoter regions.The read count density was large at 1000 bp upstream and downstream of the transcription start site,implying the role of BmJun in regulating downstream genes.GO functional enrichment analysis revealed that the BmJun bound genes were involved in a number of functional classes,including tissue and organ development,cytoplasmic membrane composition,signaling receptor activity,molecular sensor activity,and transmembrane signaling receptor activity.KEGG pathway enrichment results showed that BmJun binding-related genes were significantly enriched in Wnt signaling pathway,insect hormone biosynthesis,metabolism of xenobiotics by cytochrome P450,retinol metabolism,Hippo signaling pathway,melanogenesis,biosynthesis of unsaturated fatty acids,cAMP signaling pathway,mTOR signaling pathway,and apoptosis pathway,etc.Finally,the target genes bound by BmJun on the host genome were obtained and identified,and the downstream regulatory network of BmJun was analyzed based on the reported functions of Jun.This study will provide a new reference for in-depth research on the regulation of related functions by the silkworm transcription factor BmJun during BmNPV infection.
