摘要
目的采用UPLC-Q-TOF技术,通过非靶向代谢组学研究方法探究肝缺血再灌注模型中代谢物的变化。方法将14只小鼠(C57BL/6J)随机分为两组,分别为假手术组(Sham)和手术组(I/R),构建肝脏缺血再灌注损伤模型。检测小鼠血清中AST和ALT的水平。利用超高效液相-四极杆飞行时间质谱仪(UPLC-Q-TOF)采集小鼠血清样品代谢物信息,采用PCA法、OPLS-DA法筛选差异代谢物,并对代谢通路进行富集分析。结果假手术组和手术组比较,筛选出花生四烯酸等13种差异代谢物,主要涉及花生四烯酸代谢、类固醇激素生物合成等代谢通路。与假手术组相比,手术组小鼠血清中ALT和AST水平明显升高。结论肝脏缺血再灌注的病理状态可造成小鼠血清中代谢物谱的显著变化,其中脂类和类脂类化合物占比最高。本研究从非靶向代谢组学角度揭示了肝脏缺血再灌注病理模型中潜在的13个生物标记物,为肝损伤早期干预和临床药物研发提供参考。
Objective To explore the changes of metabolites in hepatic ischemia-reperfusion injury model by using UPLC-Q-TOF technology and non-targeted metabolomics research methods.Methods Fourteen mice(C57BL/6J)were randomly divided into two groups:sham group(Sham)and operation group(I/R).A hepatic ischemia-reperfusion injury model was established.The levels of AST and ALT in mouse serum were detected.The metabolic information of mouse serum samples was collected by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF).PCA and OPLS-DA methods were used to screen differential metabolites,and metabolic pathway enrichment analysis was performed.Results Thirteen differential metabolites,including arachidonic acid,were screened out by comparing the Sham group and the I/R group.They were mainly involved in arachidonic acid metabolism and steroid hormone biosynthesis pathways.Compared with the sham operation group,the levels of ALT and AST in the serum of the I/R group mice were significantly increased.Conclusion The pathological state of liver ischemia-reperfusion can cause significant changes in the metabolite profile of mouse serum,with lipids and lipid-like compounds accounting for the highest proportion.This study revealed 13 potential biomarkers in the hepatic ischemia-reperfusion pathological model from the perspective of non-targeted metabolomics,providing a reference for early intervention of liver injury and clinical drug development.
