黑果枸杞花青素调控低氧下八眉三元猪心脏抗氧化/炎症/免疫功能及Nrf2通路的作用机制

The regulatory mechanism of Lycium ruthenicum anthocyanin on antioxidant/inflammatory/immune function and the Nrf2 pathway of the hearts of Bamei ternary hybrid pigs under hypoxic conditions

摘要 本研究旨在探究黑果枸杞花青素对低氧环境下八眉三元猪血清抗氧化、炎症和免疫功能的调控作用及其通过Nrf2通路、低氧适应因子对心肌氧化应激的缓解作用。通过向日粮添加不同水平的黑果枸杞花青素(5、10、15 g/kg)饲养八眉三元猪并采集其血液、心脏组织及心肌细胞,检测其心脏血清抗氧化指标、炎性因子及免疫球蛋白水平;通过RT-qPCR、Western-blot技术分析体外低氧心肌细胞模型及八眉三元猪心肌组织Nrf2通路基因及低氧适应因子的mRNA和蛋白表达。结果显示,低氧环境显著降低血清GSH-Px、CAT、SOD活性及T-AOC(P<0.05),提高MDA含量(P<0.05),并诱发炎症(TNF-α、IL-6上升,IL-10下降)与免疫功能抑制(IgA、IgG、IgM减少);同时抑制心肌Nrf2通路相关因子Nrf2、HO-1、NQO1、KAP1的mRNA及蛋白表达;低氧适应相关因子HIF-1α、VEGF蛋白下调,PHD2上升。补充黑果枸杞花青素(10 g/kg剂量组效果最佳)可显著提高GSH-Px、CAT、SOD活性及T-AOC水平(P<0.05),降低MDA含量(P<0.05),降低促炎因子TNF-α(P<0.01)和IL-6(P<0.05)水平,提高IL-10水平(P<0.05);并显著增加IgA、IgG和IgM含量(P<0.05);此外,黑果枸杞花青素10 g/kg剂量组极显著上调心肌Nrf2、HO-1、NQO1的mRNA及蛋白表达(P<0.01),促进HIF-1α和VEGF蛋白表达(P<0.05),抑制PHD2(P<0.01)。结果表明,黑果枸杞花青素可通过激活Nrf2信号通路及调节低氧适应因子表达,有效缓解低氧诱导的氧化应激,改善八眉三元猪的抗氧化能力、炎症状态和免疫功能,其中10 g/kg添加剂量效果最优,为高原畜牧低氧应激防控提供了天然干预策略。 This study aimed to investigate the regulatory effects of Lycium ruthenicum anthocyanins on serum antioxidant,inflammatory,and immune functions in Bamei ternary hybrid pigs under hypoxic conditions,as well as their alleviating effects on myocardial oxidative stress through the Nrf2 pathway and hypoxic adaptation factors.L.ruthenicum anthocyanins were added to the diet at different concentrations(5,10,15 g/kg)to feed the Bamei ternary hybrid pigs,and their blood,heart tissue,and myocardial cells were collected.The antioxidant indicators,inflammatory factors,and immunoglobulin levels in the heart serum were detected.Using RT-qPCR and Western-blot,analyse mRNA and protein expression of Nrf2 pathway genes and hypoxia-adaptive factors in an in vitro hypoxic cardiomyocyte model and in the myocardial tissue of the Bamei ternary hybrid pigs.The results showed that hypoxia significantly reduced serum GSH-Px,CAT,and SOD activity and T-AOC(P<0.05),increased MDA content(P<0.05),and induced inflammatory responses(increased TNF-α and IL-6,decreased IL-10)and immune function suppression(decreased IgA,IgG,and IgM),simultaneously inhibiting the expression of Nrf2,HO-1,NQ01,and KAP1 mRNA and protein levels related to the myocardial Nrf2 pathway.Downregulated HIF-1α and VEGF protein expression,while increasing PHD2 expression.Supplementation with L.ruthenicum anthocyanins(with the 10 g/kg dose group showing the best effect)significantly increased GSH-Px,CAT,SOD activity,and T-AOC levels(P<0.05),reduced MDA content(P<0.05),decreased pro-inflammatory factors TNF-α(P<0.01)and IL-6(P<0.05)levels,and increased IL-10 levels(P<0.05),and significantly increased IgA,IgG,and IgM content(P<0.05).Additionally,this group significantly upregulated the mRNA and protein expression of myocardial Nrf2,HO-1,and NQ01(P<0.01),promoted HIF-1α and VEGF protein expression(P<0.05),and inhibited PHD2(P<0.01).L.ruthenicum anthocyanins can effectively alleviate hypoxia-induced oxidative stress by activating the Nrf2 signalling pathway and regulating the expression of hypoxia adaptation factors,thereby improving the antioxidant capacity,inflammatory status,and immune function of Bamei ternary hybrid pigs.The optimal effect was observed at a dosage of 10 g/kg,providing a natural intervention strategy for hypoxia stress prevention and control in high-altitude livestock farming.
