摘要
目的:探讨积雪草苷对环磷酸腺苷(cAMP)/蛋白激酶A(PKA)信号通路及支气管哮喘(BA)大鼠气道炎症的影响。方法:卵清蛋白(OVA)致敏法建立BA大鼠模型,随机分为模型组、积雪草苷低、高剂量组、积雪草苷高剂量+SQ22536(cAMP抑制剂)组,另取10只大鼠设为对照组,以积雪草苷和SQ22536分组处理后,肺功能检测仪检测大鼠肺功能,HE染色检测肺组织病理,PAS染色检测肺组织黏液分泌,吉姆萨染色和全自动血液生化分析仪检测支气管肺泡灌洗液(BALF)和外周血中炎症细胞数,ELISA检测BALF和血清中炎症因子(IL-17、IL-6)水平及肺组织cAMP水平,Western blot检测肺组织cAMP/PKA通路蛋白表达。结果:BA大鼠模型制备成功并诱发气道炎症;积雪草苷降低大鼠气道炎症评分、酸性黏液阳性比、BALF和外周血中白细胞数、嗜酸性粒细胞数、IL-17及IL-6水平,升高每分钟通气量(MV)、用力肺活量(FVC)、呼气峰流值(PEF)、肺组织cAMP水平及p-PKA/PKA(P<0.05);SQ22536可减弱积雪草苷对BA大鼠各炎症指标的作用。结论:积雪草苷可通过激活cAMP/PKA信号抑制BA大鼠炎症细胞及炎症因子产生,从而减轻气道炎症及肺组织损伤,改善肺功能,其抗炎作用与使用剂量相关。
Objective:To investigate effects of asiaticoside on cyclic adenosine monophosphate(cAMP)/protein kinase A(PKA)signaling pathway and airway inflammation in rats with bronchial asthma(BA).Methods:A BA rat model was established using ovalbumin(OVA)sensitization method,rats were randomly separated into model group,low-dose,high-dose asiaticoside groups,high-dose asiaticoside+SQ22536(cAMP inhibitor)group,another 10 rats were selected as control group.After grouping with asiaticoside and SQ22536,pulmonary function was detected by pulmonary function detector,lung tissue pathology was detected by HE staining,mucous secretion of lung tissue was detected by PAS staining,number of inflammatory cells in peripheral broncho alveolar lavage fluid(BALF)and peripheral blood were detected by Giemsa staining and automatic blood biochemical analyzer.Inflammatory factors(IL-17 and IL-6)in serum and BALF and cAMP levels in lung tissue were detected by ELISA.cAMP/PKA pathway protein expression in lung tissue was detected by Western blot.Results:BA rat model was successfully prepared and induced airway inflammation;asiaticoside decreased airway inflammation score,positive ratio of acidic mucus,levels of white blood cells,eosinophils,IL-17 and IL-6 in BALF and peripheral blood,and increased levels of minute ventilation volume(MV),forced vital capacity(FVC),peak expiratory flow(PEF),cAMP in lung tissue and p-PKA/PKA(P<0.05);SQ22536 could weaken effects of asiaticoside on various inflammatory indexes in BA rats.Conclusion:Asiaticoside can inhibit production of inflammatory cells and cytokines in BA rats by activating cAMP/PKA signal,thereby reducing airway inflammation and lung tissue damage,and improving lung function,and its anti-inflammatory effect is related to dosage used.
