壮药火钱胶囊对D-GalN/LPS所致急性肝衰竭小鼠的保护作用

摘要 目的研究壮药火钱胶囊对D-氨基半乳糖/脂多糖(D-GalN/LPS)所致急性肝衰竭小鼠的保护作用。方法小鼠随机分为正常组、模型组、水飞蓟宾组(100 mg/kg)和火钱胶囊低、中、高剂量组(365、730、1460 mg/kg),每组25只。预防性灌胃给药10 d后,采用单次腹腔注射给予D-GalN(700 mg/kg)和LPS(10μg/kg)建立肝衰竭模型。检测小鼠存活率;计算肝指数;检测血清AST、ALT、IL-1β、IL-6、TNF-α、MCP-1水平,肝组织MDA、NO、IL-1β、IL-6、TNF-α水平和SOD活性;HE染色观察肝组织病理学改变,RT-qPCR法检测肝组织IL-1β、IL-6、TNF-α、iNOS、NF-κB、IκBα、TLR 4 mRNA表达;Western blot法检测肝组织Bax、Bcl-2、Bcl-xL、p-p38、p-p65蛋白表达。结果与模型组比较,水飞蓟宾组和火钱胶囊各剂量组小鼠存活率提高,肝指数、血清AST、ALT、IL-1β、IL-6、TNF-α、MCP-1和肝组织MDA、NO、IL-1β、IL-6、TNF-α水平降低(P<0.05,P<0.01),肝组织SOD活性升高(P<0.01),肝组织病理损伤减轻;水飞蓟宾组和火钱胶囊高剂量组小鼠肝组织IL-1β、IL-6、TNF-α、iNOS、NF-κB、IκBα、TLR 4 mRNA和Bax、p-p38、p-p65蛋白表达降低(P<0.05,P<0.01)。结论火钱胶囊对D-GalN/LPS所致急性肝衰竭小鼠具有保护作用,其机制可能与抑制肝细胞炎症和凋亡有关。 AIM To investigate the protective effects of Zhuang-medicine Huoqian Capsules on acute liver failure(ALF)induced by D-galactosamine/lipopolysaccharide(D-GalN/LPS)in mice.METHODS The mice were randomly divided into the normal group,the model group,the silibinin group(100 mg/kg)and the low-dose,medium-dose and high-dose Huoqian Capsules groups(365,730,1460 mg/kg),with 25 mice in each group.After 10 days preventive intragastric administration of the corresponding drug,single intraperitoneal injection of D-GalN(700 mg/kg)and LPS(10μg/kg)was conducted to establish the mouse model of ALF.The mice had their survival rate examined;their liver index calculated;their serum levels of AST,ALT,IL-1β,IL-6,TNF-α and MCP-1 and hepatic levels of MDA,NO,IL-1β,IL-6 and TNF-α and SOD activity measured;their liver histopathological changed examined by HE staining;their hepatic mRNA expressions of IL-1β,IL-6,TNF-α,iNOS,NF-κB,IκBαand TLR 4 measured by RT-qPCR;and their hepatic protein expressions of Bax,Bcl-2,Bcl-xL,p-p38 and p-p65 measured by Western blot.RESULTS Compared with the model group,the groups intervened with either silibinin or Huoqian Capsules displayed increased survival rate;decreased levels of liver index,serum AST,ALT,IL-1β,IL-6,TNF-α,MCP-1 and hepatic MDA,NO,IL-1β,IL-6 and TNF-α(P<0.05,P<0.01);increased hepatic SOD activity(P<0.01);and alleviated pathological damage of the liver tissue.Additionally,the silibinin and high-dose Huoqian Capsules groups shared decreased hepatic expressions of IL-1β,IL-6,TNF-α,iNOS,NF-κB,IκBα,TLR 4 mRNA and Bax,p-p38,p-p65 proteins(P<0.05,P<0.01).CONCLUSION With their good hepatoprotective effects on the D-GalN/LPS-induced mouse model of ALF,Huoqian Capsules may inhibit the hepatocytic inflammation and apoptosis.
