摘要
本研究通过建立Walker-256大鼠癌性腹水模型,观察甘遂醋炙前后对癌性腹水模型大鼠逐水利尿的影响,从而探究甘遂经醋炙后是否具有更好的逐水利尿功效。通过腹腔注射Walker-256细胞株构建癌性腹水大鼠模型,观察不同剂量的生甘遂、醋甘遂以及阳性药呋塞米对大鼠尿量、腹水量、水通道蛋白含量、肾素血管紧张素醛固酮系统(RAAS)以及尿液离子浓度的影响。结果表明,与模型组比较,醋甘遂组尿量显著升高,腹水量显著降低,尿液上清液中的离子浓度明显降低,血清中的ALD、ADH浓度也显著降低,与模型组比较,各给药组水通道蛋白AQP1、AQP8、AQP2显著降低。生甘遂和醋甘遂都有一定利尿功效,且能抑制RAAS系统,改善电解质失调的情况;同时也能够调节大鼠体内水通道蛋白,从而控制癌性腹水产生,缓解了大鼠肝肾组织的损伤。在相同用药剂量下,与生甘遂相比,醋甘遂更明显地发挥了抑制腹水生成、利尿、调节RAAS系统、调节水通道蛋白以及调节离子紊乱,在治疗癌性腹水方面具有优势。因此,本研究可为甘遂进一步的临床应用提供理论依据。
through the establishment of Walker-256 cancerous ascites model in rats,we observed the Euphorbia kansui vinegar before and after acupuncture on cancerous ascites model rats by water diuretic effect,so as to explore the Euphorbia kansui has better by water after vinegar main diuretic effects.The rat model of cancer ascites was established by intraperitoneal injection of Walter-256 cell line.The effects of different doses of raw Euphorbia kansui,acehansui and positive drug furosemide on urine volume,ascites volume,aquaporin content,renin-angiotensin-aldosterone system(RAAS)and urine ion concentration were observed.Compared with the model group,the urine volume was significantly increased,the ascites volume was significantly decreased,the ion concentration in urine supernatant was significantly decreased,and the ALD and ADH concentrations in serum were also significantly decreased in the vinegar Euphorbia kansui group.Compared with the model group,the aquaporins AQP1,AQP8,and AQP2 were significantly decreased in each drug group.Both raw Euphorbia kansui and vinegar Euphorbia kansui have diuretic effect and can inhibit the RAAS system and improve electrolyte imbalance.At the same time,it can regulate aquaporins in rats,thereby controlling the production of cancerous ascites and alleviate the damage of liver and kidney tissue in rats.At the same dose,compared with raw Euphorbia kansui,vinegar Euphorbia kansui has more obvious effects on inhibiting ascites formation,diuresis,regulating RAAS system,aquaporin and ion disorder,and has advantages in the treatment of cancer ascites.Therefore,this study provides a theoretical basis for the further clinical application of Euphorbia kansui.
