蒲公英调节PPAR信号通路对2型糖尿病大鼠糖脂代谢的影响

Effects of Pugongying(Taraxaci Herba)on the Disorder of Glucose and Lipid Metabolism in Type 2 Diabetes Rats by Regulating PPAR Signaling Pathway

摘要 目的旨在探讨蒲公英对2型糖尿病(T2DM)大鼠糖脂代谢的影响,并揭示其潜在机制,特别是通过激活PPAR信号通路改善代谢紊乱的作用。方法采用高脂喂养及注射链脲佐菌素的方法构建T2DM大鼠模型,随机分为对照组、模型组和蒲公英组(400 mg/kg)。通过高效液相色谱法分析蒲公英成分,通过肝脏转录组学分析差异基因、进行GO和KEGG通路富集分析,结合肝脏组织样本的HE染色进行病理观察,通过实时荧光定量逆转录聚合酶链式反应(qRT-PCR)和蛋白免疫印迹法(Western blot)检测Acsl3、Me1、Plin2的基因和蛋白表达。结果结果表明,蒲公英显著降低了糖尿病大鼠的空腹血糖(FBG)和葡萄糖耐量,改善了血脂水平,降低了炎症因子[如肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)]的水平。此外,蒲公英处理C肽和胰岛素的浓度显著提高,表明其改善了胰岛β细胞功能。HE染色结果显示肝组织病理学特征有所改善,转录组学分析显示,蒲公英组中与PPAR信号通路相关的基因明显上调。Western blot结果表明蒲公英组肝脏Acsl3、Me1、Plin2蛋白相对表达量增加,qRT-PCR结果表明蒲公英组Acsl3、Me1、Plin2转录表达增加。结论蒲公英通过激活PPAR信号通路和上调相关基因的表达,显著改善了T2DM大鼠的糖脂代谢紊乱,为蒲公英辅助治疗T2DM提供了实验证据。研究结果强调了蒲公英在糖尿病管理中的应用前景,提出其作为自然疗法的可能性。 Objective This study aims to investigate the effects of Pugongying(Taraxaci Herba)on glucose and lipid metabolism in rats with type 2 diabetes mellitus(T2DM)and to elucidate its underlying mechanisms,particularly the role of activating the PPAR signaling pathway in improving metabolic disorders.Methods A T2DM rat model was established using a high-fat diet and streptozotocin injection.The rats were randomly divided into three groups:control group,model groip,and Dandelion group,with the Dandelion group receiving a dosage of 400 mg/kg.The components of Pugongying(Taraxaci Herba)were analyzed using high-performance liquid chromatography.The differential gene expression in liver tissues was assessed through transcriptomic analysis,followed by GO and KEGG enrichment analyses.Histopathological observations were conducted on liver tissue samples stained with hematoxylin and eosin(HE).Gene and protein expression levels were evaluated using quantitative real-time reverse transcription polymerase chain reaction(qRT-PCR)and Western blotting.Results The results demonstrated that Pugongying(Taraxaci Herba)significantly reduced fasting blood glucose(FBG)and improved glucose tolerance in diabetic rats,along with enhancements in lipid profiles and reductions in inflammatory cytokines(such as TNF-αand IL-1β).Furthermore,the concentrations of C-peptide and insulin were markedly elevated in the Dandelion treatment group,indicating an improvement inβ-cell function.HE staining results showed histopathological improvements,and transcriptomic analysis revealed a significant upregulation of genes associated with the PPAR signaling pathway in the Dandelion group.Western blot analyses indicated increased relative expression levels of Acsl3,Me1,and Plin2 proteins in the liver of the Dandelion group,while qRT-PCR results showed an increase in the transcription of Acsl3,Me1,and Plin2.Conclusion Pugongying(Taraxaci Herba)significantly ameliorates glucose and lipid metabolic disorders in T2DM rats by activating the PPAR signaling pathway and enhancing the expression of related genes.This provides experimental evidence for the potential of dandelion as an adjunct therapy for T2DM.The findings underscore the prospects of Pugongying(Taraxaci Herba)in diabetes management,suggesting its potential as a natural therapeutic agent.
