摘要
目的 探讨短链脂肪酸(short chain fatty acids, SCFAs)在食物过敏中的作用及调控机制,为预防和治疗食物过敏提供策略。方法 采用α-酪蛋白建立食物过敏细胞和动物模型,并给予SCFAs干预。RT-qPCR和Western blotting法检测细胞炎症因子及信号通路关键基因mRNA和蛋白表达水平。HE染色和阿尔辛蓝-过碘酸雪夫染色(alcian blue-periodic acid-Schiff staining, AB-PAS)检测过敏小鼠结肠组织的病理及杯状细胞改变。结果 SCFAs干预可显著下调α-酪蛋白刺激下人正常结肠上皮细胞(HCoEpiC)和小鼠结肠组织内促炎性细胞因子IL-1β、IL-6、TNF-α、COX-2 mRNA水平,并抑制HCoEpiC细胞内哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin, mTOR)表达。HE染色和AB-PAS染色结果表明,食物过敏小鼠结肠组织内存在多处病理损伤,SCFAs干预可显著缓解食物过敏诱发的结肠组织损伤,增加杯状细胞数量。Western blotting结果显示,SCFAs可显著抑制过敏小鼠结肠组织内Janus激酶(Janus kinase, JAK)、转录激活因子3(signal transducer and activator of transcription 3,STAT3)和mTOR的磷酸化以及COX-2蛋白的表达水平。结论 SCFAs通过STAT3抑制肠上皮细胞炎性细胞因子的表达,从而抑制食物过敏诱发的肠道炎症反应。
Objective To explore the role and regulatory mechanism of short chain fatty acids(SCFAs)in food allergy,and to provide strategies for the prevention and treatment of food allergy.Methods Food allergic cells and animal models were established usingα-casein and given SCFAs intervention.The mRNA and protein expression levels of inflammatory factors and the key genes of signaling pathway were detected by RT-qPCR and Western blotting.HE staining and AB-PAS staining were used to detect of pathological changes and alterations in goblet cells in the colon tissue of allergic mice.Results SCFAs intervention can significantly down-regulate the mRNA levels of pro-inflammatory cytokines IL-1β,IL-6,TNF-αand COX-2 in HCoEpiC cells and mice colon tissues stimulated byα-casein,and inhibit the expression of mTOR in HCoEpiC cells.The results of HE and AB-PAS staining showed that there were many pathological lesions in colon tissue of CMPA mice,and SCFAs intervention could significantly alleviate the food allergy induced colon tissue injury and increase the number of goblet cells.Western blotting results showed that SCFAs significantly inhibited the phosphorylation of JAK,STAT3,mTOR and the expression of COX-2 protein in the colon tissue of allergic mice.Conclusion SCFAs inhibit the expression of inflammatory cytokines in epithelial cells through STAT3,thereby inhibiting food allergy induced intestinal inflammation.
