栝楼茎叶的解暑功能评价及功能性饮料研制

Evaluation of the Summerheat-clearing Effect of Trichosanthes kirilowii Stems and Leaves and Development of A Functional Beverage

摘要 目的:评价栝楼茎叶解暑活性,并开发栝楼茎叶功能性饮料。方法:采用高温高湿气候舱加热的方法制备中暑小鼠模型,在造模前灌胃栝楼茎叶提取物12 d,对栝楼茎叶解暑活性进行评价。以栝楼茎叶为原料,感官评价(色泽、气味、口感、组织形态)为指标,通过单因素试验及正交试验对栝楼茎叶功能性饮料配方进行优化,采用福林酚法、NaNO2-Al(NO3)3-NaOH法、苯酚-硫酸法、咔唑-硫酸法、双缩脲反应分别测定栝楼茎叶功能性饮料中的总酚、总黄酮、中性多糖、酸性多糖及蛋白质含量,利用1,1-二硝基-2-三硝基苯肼(DPPH)自由基清除率、2,2'-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)自由基清除率、羟基自由基清除率和总还原能力评价其抗氧化活性。结果:栝楼茎叶可改善中暑模型小鼠的肝脏指数,调节纤维蛋白原(FIB)水平,延长血浆凝血酶原时间(PT),降低乳酸脱氢酶(LDH)活性,从而改善凝血功能及心功能。栝楼茎叶功能性饮料最优配方为栝楼茎叶添加量10%,复配甜味剂添加量2%,柠檬酸添加量0.05%。该饮料每100 mL中含有总酚(19.59±0.05)mg、总黄酮(3.60±0.48)mg、中性多糖(171.13±3.99)mg、酸性多糖(98.43±2.06)mg及蛋白质(527.52±9.19)mg。清除ABTS自由基、DPPH自由基、羟基自由基的半数抑制浓度(IC50)分别为(0.25±0.04)、(0.82±0.10)、(5.73±0.55)mg·mL^(-1)。结论:栝楼茎叶具有解暑功能,制备的栝楼茎叶功能性饮料澄清,呈明亮的棕黄色,带有栝楼茎叶独特的清香,酸甜可口,且具有抗氧化能力。 Objective:To evaluate the summerheat-clearing activity of Trichosanthes kirilowii stems and leaves and develop a functional beverage with T.kirilowii stem and leaf extract(TSLE).Methods:A mouse model of heatstroke was established by exposure to a high-temperature and high-humidity climate chamber.Mice were orally administered with TSLE for 12 days prior to modeling to evaluate the protective effect of TSLE.The formula of the functional beverage developed with TSLE was optimized via single-factor and orthogonal experiments based on sensory evaluation of the color,odor,taste,and texture.The content of total phenols,total flavonoids,neutral polysaccharides,acidic polysaccharides,and proteins in the functional beverage were determined by the Folin-phenol method,NaNO2-Al(NO3)3-NaOH method,phenolsulfuric acid method,carbazole-sulfuric acid method,and biuret reaction,respectively.The antioxidant activities were evaluated based on the 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging rate,2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid,ABTS)radical scavenging rate,hydroxyl radical scavenging rate,and total reducing capacity.Results:TSLE ameliorated the liver index,regulated the fibrinogen(FIB)level,prolonged the plasma prothrombin time(PT),and reduced the lactate dehydrogenase(LDH)activity,thereby improving the coagulation function and cardiac function in the mouse model of heatstroke.The optimal beverage formula was determined as follows:10%TSLE,2%compound sweetener,and 0.05%citric acid.The beverage(per 100 mL)contained(19.59±0.05)mg total phenols,(3.60±0.48)mg total flavonoids,(171.13±3.99)mg neutral polysaccharides,(98.43±2.06)mg acid polysaccharides,and(527.52±9.19)mg proteins.The median inhibitory concentration(IC50)values of the beverage for scavenging ABTS radicals,DPPH radicals,and hydroxyl radicals were(0.25±0.04),(0.82±0.10),and(5.73±0.55)mg·mL^(-1),respectively.Conclusion:T.kirilowii stems and leaves possess significant summerheat-clearing properties.The prepared TSLE functional beverage presents a bright brownishyellow color,has the characteristic arom of T.kirilowii stems and leaves,a clear texture,a sweet-sour balanced taste,and antioxidant properties.
