布氏白僵菌菌株JG-17侵染对华北大黑鳃金龟、暗黑鳃金龟和铜绿丽金龟幼虫体内抗氧化酶和解毒酶活性的影响

Effects of Beauveria brongniartii strain JG-17 infection on the activities of antioxidant and detoxification enzymes in larvae of scarab beetles Holotrichia oblita,H.parallela and Anomala corpulenta(Coleoptera:Scarabaeidae)

摘要 【目的】探究病原体侵染后华北大黑鳃金龟Holotrichia oblita、暗黑鳃金龟H.parallela和铜绿丽金龟Anomala corpulenta体内抗氧化酶和解毒酶活性的响应机制。【方法】利用布氏白僵菌Beauveria brongniartii菌株JG-17孢悬液(1×10^(8)孢子/mL)分别侵染华北大黑鳃金龟、暗黑鳃金龟和铜绿丽金龟2龄幼虫,侵染后1,3,5,7,9和11 d测定3种金龟子幼虫体内的总蛋白含量、抗氧化酶[超氧化物歧化酶(superoxide dismutase,SOD)、过氧化物酶(peroxidase,POD)和过氧化氢酶(catalase,CAT)]及解毒酶[羧酸酯酶(carboxylesterase,CarE)、谷胱甘肽S-转移酶(glutathione S-transferase,GST)和乙酰胆碱酯酶(acetylcholinesterase,AChE)]的活性变化。【结果】布氏白僵菌菌株JG-17侵染后不同天数,3种金龟子幼虫体内的SOD活性均被激活,华北大黑鳃金龟幼虫体内的SOD活性的响应速度要快于暗黑鳃金龟和铜绿丽金龟幼虫体内的SOD活性;3种金龟子幼虫体内的CAT活性均表现为先升高后降低,华北大黑鳃金龟、暗黑鳃金龟和铜绿丽金龟幼虫体内的CAT活性分别在侵染后7,7和9 d达到高峰;3种被侵染的金龟子幼虫体内的POD活性的变化趋势存在差异,暗黑鳃金龟幼虫体内的POD活性在布氏白僵菌菌株JG-17侵染后1-7 d缓慢上升,侵染后9 d显著降低,华北大黑鳃金龟幼虫体内的POD活性呈“M”型变化趋势,铜绿丽金龟幼虫体内的POD活性在侵染后9和11 d显著升高。布氏白僵菌菌株JG-17侵染后,3种金龟子幼虫体内的GST活性均表现为先升高后降低,暗黑鳃金龟、华北大黑鳃金龟和铜绿丽金龟幼虫体内的GST活性分别在侵染后3,5和7 d显著升高,侵染后5,7和9 d达到峰值后下降。布氏白僵菌菌株JG-17侵染后,暗黑鳃金龟幼虫体内的CarE活性持续下降,华北大黑鳃金龟幼虫体内的CarE活性先升高后降低,铜绿丽金龟幼虫体内的CarE活性在侵染后7-11 d持续升高;3种被侵染的金龟子幼虫体内的AChE活性均表现为先升高后降低,其中铜绿丽金龟幼虫体内的AChE活性及其变化幅度均高于暗黑鳃金龟和华北大黑鳃金龟。【结论】本研究明确了3种金龟子防御白僵菌侵染时酶活性的响应机制,对揭示昆虫免疫响应以及研制高效微生物杀虫剂具有重要意义。 【Aim】To explore the response mechanism of antioxidant and detoxification enzyme activities in Holotrichia oblita,H.parallela and Anomala corpulenta following pathogen infection.【Methods】The 2nd instar larvae of H.oblita,H.parallela and A.corpulenta were infected by the spore suspension of Beauveria brongniartii strain JG-17(1×10^(8)spores/mL),and the total protein content and the activities of the antioxidant enzymes including superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT),and the detoxification enzymes including carboxylesterase(CarE),glutathione S-transferase(GST)and acetylcholinesterase(AChE)in larvae of the three scarab species were determined at 1,3,5,7,9 and 11 d post infection(dpi).【Results】The SOD activities in larvae of the three scarab species infected by B.brongniartii strain JG-17 were activated at different dpi,with the SOD activity in H.oblita larvae responding faster than those in H.parallela and A.corpulenta larvae.The CAT activities in larvae of the three scarab species infected by B.brongniartii strain JG-17 showed a pattern of initial increase and subsequent decrease,with the CAT activity peaks in H.oblita,H.parallela and A.corpulenta larvae at 7,7 and 9 dpi,respectively.These three infected scarab species displayed different change trends in POD activity.The POD activity in the infected H.parallela larvae by B.brongniartii strain JG-17 increased slowly from 1 to 7 dpi,and decreased significantly at 9 dpi,that in the infected H.oblita larvae showed an“M”-shaped change trend,and that in the infected A.corpulenta larvae increased significantly at 9 and 11 dpi.After infection by B.brongniartii strain JG-17,the GST activity showed a pattern of initial increase and subsequent decrease in the three infected scarab species.The GST activities in the infected H.oblita,H.parallela and A.corpulenta larvae increased significantly at 3,5 and 7 dpi,respectively,while reached the peak at 5,7 and 9 dpi,respectively,followed by decrease.After infection by B.brongniartii strain JG-17,the CarE activity decreased continuously in the infected H.parallela larvae throughout the experimental process,while displayed decrease following increase in H.oblita larvae.And the CarE activity in the infected A.corpulenta larvae increased continuously from 7 to 11 dpi.The AChE activity showed increase followed by decrease in larvae of the three scarab species infected by B.brongniartii strain JG-17,and the AChE activity and its change degree were higher in A.corpulenta larvae than in H.oblita and H.parallela larvae.【Conclusion】Altogether,these results indicate the response mechanisms of enzymatic activities in these three scarab species against B.bassiana infection,providing valuable insights into insect immune responses and supporting the development of high-efficacy microbial insecticides.
