大白菜抗TuMV近等基因系的分子标记辅助选育

摘要 为探究大白菜芜菁花叶病毒病(Turnip mosaic virus,TuMV)基因TuRBCS01的功能,丰富大白菜种质资源,以大白菜感TuMV的骨干亲本材料‘冠291’为轮回亲本,TuMV抗源材料‘8407’为供体亲本,利用BrID10723和mBr4055为前景选择标记,49对在两亲本间多态性好、染色体分布相对均匀的InDel或SSR共显性标记为背景标记,构建抗TuMV的‘冠291’近等基因系。结果表明:1)BC_(2)代背景恢复率最高达94.44%,BC_(3)F_(1)代背景恢复率最高为97.96%;2)在BC_(3)F_(2)代获得全部恢复‘冠291’基因组的单株,再经过一代自交筛选,可获得纯合抗TuMV‘冠291’近等基因系;3)部分近等基因系的结球相关性状与‘冠291’无显著差异。这些近等基因系的获得为TuRBCS01基因的克隆及功能研究奠定了基础,同时为大白菜抗TuMV新品种的培育提供了优良种质材料。 To explore the function of Turnip mosaic virus gene TuRBCS01 in Chinese cabbage,and to enrich the germplasm resources,the TuMV resistant‘Guan 291’near-isogenic line was constructed using 49 pairs of InDel or SSR codominant markers as the background makers which with good polymorphism and relatively uniform distribution on chromosomes,BrID10723 and mBr4055 as the foreground selection markers,Chinese cabbage TuMV susceptible backbone parental material‘Guan 291’as the reincarnation parent,TuMV-resistant source material‘8407’as the donor parent.The results showed that 1)the highest background recovery rate of BC_(2)generation reached 94.44%,the highest background recovery rate of BC_(3)F_(1)generation was 97.96%;2)In the BC_(3)F_(2)generation,individuals could be obtained that fully recover the‘Guan 291’genome.After one more generation of self-pollination selection,a homozygous TuMV-resistant‘Guan 291’near-isogenic line could be obtained;3)No significant difference in bolting-related traits existed compared‘Guan 291’to some near isogenic lines.The acquisition of these near-isogenic lines has laid the foundation for the cloning and functional study of the gene TuRBCS01.At the same time,it provides excellent germplasm materials for breeding of new TuMV resistant varieties in Chinese cabbage.
