虎杖苷调节HIF-1α/VEGF信号通路对缺血性脑卒中大鼠神经损伤的影响

The Effect of Polydatin on the Nerve Injury in Rats with Ischemic Stroke through Regulating the HIF-1α/VEGF Signaling Pathway

摘要 目的:探讨虎杖苷调节低氧诱导因子-1α(HIF-1α)/血管内皮生长因子(VEGF)信号通路对缺血性脑卒中大鼠神经损伤的影响。方法:通过线栓法建立缺血性脑卒中大鼠模型,将造模后的大鼠随机分为模型组、虎杖苷组(10 mg/kg虎杖苷)、虎杖苷+YC-1组(10 mg/kg虎杖苷+4 mg/kg YC-1)、阳性药物组(1.0 mg/kg尼莫地平),并以不插入线栓的10只大鼠为假手术组,干预结束后,评估大鼠神经行为、梗死面积;苏木精-伊红(HE)、尼氏染色检测脑组织形态学变化以及神经元的数量变化;免疫组化法测定脑组织中微血管密度(CD31)表达;蛋白免疫印迹法(Western Blot)检测脑组织中HIF-1α、VEGF蛋白表达。结果:与假手术组相比,模型组神经行为评分、梗死面积明显增加,神经元数量、CD31阳性表达、HIF-1α、VEGF蛋白表达明显降低,差异均有统计学意义(P<0.05);与模型组相比,虎杖苷组、阳性药物组神经行为评分、梗死面积明显降低,神经元数量、CD31阳性表达、HIF-1α、VEGF蛋白表达明显增加,差异均有统计学意义(P<0.05);与虎杖苷组相比,虎杖苷+YC-1组神经行为评分、梗死面积明显增加,神经元数量、CD31阳性表达、HIF-1α、VEGF蛋白表达明显降低,差异均有统计学意义(P<0.05)。结论:虎杖苷可以降低缺血性脑卒中大鼠神经行为评分,缩小梗死面积,增加神经元数目,减轻大鼠神经损伤,可能与调节HIF-1α/VEGF信号通路有关。 Objective:To explore the effect of polydatin on the nerve injury in rats with ischemic stroke through regulating the hypoxiainducible factor-1α(HIF-1α)/vascular endothelial growth factor(VEGF)signaling pathway.Methods:The rat model of ischemic stroke was established by the wire suture method.After modeling,the rats were randomly divided into the model group,the polydatin group(10 mg/kg polydatin),the polydatin+YC-1 group(10 mg/kg polydatin+4 mg/kg YC-1),and the positive drug group(1.0 mg/kg nimodipine),and 10 rats without wire suture insertion were set as the sham operation group.After the intervention,the neurological behaviors of the rats and the infarction area were evaluated.The hematoxylin-eosin(HE)and Nissl staining were used to detect the morphological changes of the brain tissue and the changes in the number of neurons.The expression of microvessel density(CD31)in the brain tissue was determined by immunohistochemistry.The protein expressions of HIF-1αand VEGF in the brain tissue were detected by Western Blot.Results:Compared with the sham operation group,the neurological behavior scores and infarction areas in the model group significantly increased,while the number of neurons,the positive expression of CD31,the protein expressions of HIF-1αand VEGF significantly decreased(P<0.05).Compared with the model group,the neurological behavior scores and infarction areas in the polydatin group and the positive drug group significantly decreased,while the number of neurons,the positive expression of CD31,the protein expressions of HIF-1αand VEGF significantly increased(P<0.05).Compared with the polydatin group,the neurological behavior scores and infarction areas in the polydatin+YC-1 group significantly increased,while the number of neurons,the positive expression of CD31,the protein expressions of HIF-1αand VEGF significantly decreased(P<0.05).Conclusion:Polydatin could reduce the neurological behavior score of rats with ischemic stroke,decrease the infarction area,increase the number of neurons,alleviate the nerve injury,these effects might be related to regulating the HIF-1α/VEGF signaling pathway.
