黄芪多糖对自然杀伤T细胞免疫性能调控的实验研究

摘要 目的:分析黄芪多糖(APS)对自然杀伤T细胞(NKT)免疫性能的调控作用。方法:选择健康雌性大鼠50只,通过右前腋下接种宫颈癌细胞U14构建宫颈癌大鼠模型,造模成功后随机分为对照组(n=10)、实验1组(n=20)和实验2组(n=20)。对照组予以0.9%氯化钠注射液静脉输注干预,实验1组予以不同浓度APS静脉输注干预(APS低浓度组,100 mg·kg^(-1)·d^(-1),10只;APS高浓度组,200 mg·kg^(-1)·d^(-1),10只),实验2组予以NKT细胞和不同浓度的APS静脉输注干预(APS低浓度组,100 mg·kg^(-1)·d^(-1),10只;APS高浓度组,200 mg·kg^(-1)·d^(-1),10只),均干预30 d。分析肿瘤情况、大鼠肿瘤组织CDld mRNA的表达情况,并测定大鼠肿瘤组织血清干扰素-γ(IFN-γ)、白介素(IL)-2、IL-4、肿瘤坏死因子(TNF)-α、转化生长因子-β(TGF-β)、NKT水平。结果:实验1组、实验2组干预后肿瘤体积、肿瘤质量、TNF-α和TGF-β低于对照组(P<0.05),且实验2组APS高浓度组上述指标最低(P<0.05)。干预后实验1组、实验2组抑瘤率、CDld mRNA表达水平、NKT水平及IFN-γ,IL-2,IL-4高于对照组,且实验2组APS高浓度组上述指标最高(P<0.05)。结论:黄芪多糖应用于宫颈癌治疗中可通过改善IFN-γ,IL-2,IL-4,TNF-α及TGF-β水平,调控CDld mRNA,提高NKT细胞免疫性能,抑制肿瘤进展。 Objective:To analyze the regulatory effect of astragalus polysaccharides(APS)on the immune performance of Natural Killer T cells(NKT).Methods:Fifty healthy female rats were selected,and the rat model of cervical cancer was established by inoculating U14 cells into the right anterior armpit.After successful modeling,they were randomly divided into control group(n=10),experimental group 1(n=20)and experimental group 2(n=20).The control group 1 was intervened by intravenous infusion of 0.9%sodium choride solution,and the experimental group was intervened by intravenous infusion of APS with different concentrations(APS low concentration group,100 mg·kg^(-1)·d^(-1),n=10;APS high concentration group,200 mg·kg^(-1)·d^(-1),n=10),and the experimental group 2 was intervened by intravenous infusion of NKT cells and APS with different concentrations(APS low concentration group,100 mg·kg^(-1)·d^(-1),n=10;APS high concentration group,200 mg·kg^(-1)·d^(-1),n=10),all of them were intervened for 30 days.Results:After intervention,the tumor volume,tumor weight,TNF-αand TGF-βin experimental group 1 and experimental group 2 were lower than those in control group(P<0.05),and the above indexes in experimental group 2 with high APS concentration were the lowest(P<0.05).The tumor inhibition rate,relative content of CDld mRNA,NKT level,IFN-γ,IL-2 and IL-4 in experimental group 1 and experimental group 2 were higher than those in control group,and the above indexes were the highest in experimental group 2 with high APS concentration(P<0.05).Conclusion:Astragalus polysaccharide can be used in the treatment of cervical cancer by improving the levels of IFN-γ,IL-2,IL-4,TNF-αand TGF-β,regulating CDld mRNA,improving the immune performance of NKT cells,inhibiting tumor progression.
