沙棘黄酮提取工艺优化及其生物活性研究

摘要 研究采用超声辅助醇提法提取沙棘黄酮,通过单因素试验和响应面优化,筛选得到沙棘黄酮最优工艺并从抗氧化性能和抑菌性能两方面评价沙棘黄酮的生物活性。通过单因素试验结合响应面优化分析,获取最佳工艺参数,基于此参数所得到的沙棘黄酮具有良好的体外抗氧化活性,质量浓度为120μg/mL时,对DPPH·和羟基自由基的清除率分别为99.5%、79.4%。对沙棘黄酮进行抑菌性能测定,沙棘黄酮对革兰氏阳性菌和革兰氏阴性菌均有抑制作用,对大肠杆菌、铜绿假单胞菌、枯草芽孢杆菌、金黄色葡萄球菌的MIC分别为:0.75 mg/mL、0.75 mg/mL、0.75 mg/mL、1.5 mg/mL。本试验为沙棘相关产品的精加工提供了参考。 Ultrasonic alcohol-assisted extraction of sea buckthorn flavonoids was studied.Through single factor test and response surface optimization,the optimal process of sea buckthorn flavonoids was screened and the biological activity of sea buckthorn flavonoids was evaluated from two aspects of antioxidant and antibacterial properties.The optimum process parameters were obtained by single factor test combined with response surface optimization analysis.The seabuckthorn flavonoids obtained based on this parameter had good antioxidant activity in vitro.When the mass concentration was 120μg/mL,the scavenging rates of DPPH·and hydroxyl radicals were 99.5%and 79.4%,respectively.The antibacterial properties of seabuckthorn flavonoids were determined.Seabuckthorn flavonoids had inhibitory effects on Gram-positive bacteria and Gram-negative bacteria.The MICs of Escherichia coli,Pseudomonas aeruginosa,Bacillus subtilis and Staphylococcus aureus were 0.75 mg/mL,0.75 mg/mL,0.75 mg/mL and 1.5 mg/mL,respectively.This experiment provided a reference for the finishing of sea buckthorn related products.
