基于光学成像的菌落去重复挑选技术

摘要 为解决微生物菌落培养研究中菌落挑取、培养、鉴定工作量大、重复度高的问题,提出一种基于光学成像的菌落去重复挑选策略。通过宏观成像对菌落进行计数和定位,与通过微观成像获取的菌落特征信息结合,并使用贪心算法对获得特征信息的菌落进行挑选,实现菌落挑选的去重复工作。该方法成功建立了多维度菌落识别方法与技术路线,将200个菌落样本的挑选方案的获得缩减至5 min内,且当挑菌数与菌落样本种数之比为3时,菌落覆盖率超过90%。与传统的人工去重复和连续稀释法进行菌株挑选相比,基于光学成像的菌落挑选策略能够提高菌落挑选效率,降低人力资源成本,并有效提高挑菌的覆盖率。 In order to address the issues of excessive workload and repetitive tasks involved in microbial colony culture studies,this paper proposes a strategy of colony de-duplicated selection based on optical imaging,which is to count and locate colonies through macro imaging,obtain colony characteristic information through micro imaging,and use greedy algorithm to select a few from these colonies,so as to achieve colony de-duplicated selection.The method successfully established a multi-dimensional colony identification method and technical route,and reduced the selection plans of 200 colony samples to 5 minutes while the ratio of the number of bacteria picked to the number of colony sample species was 3,the colony coverage exceeded 90%.Compared with traditional manual picking methods and serial dilution-based isolation methods,the optical imaging-based colony selection strategy enhances efficiency while reducing human resource costs,ultimately improving bacterial coverage.
