摘要
探究不同分子量和单糖组成的高良姜多糖(Alpinia officinarum Hance polysaccharides,AOHP)对乙醇诱导人胃黏膜细胞GES-1细胞损伤的保护作用.通过汤剂制备法和阴离子交换柱层析DEAE-52获得4种高良姜多糖组分(AOHP1~4),采用苯酚-硫酸法、比旋光法和高效凝胶渗透色谱法、高效液相色谱法等测定其化学组成、分子量分布和单糖组成.其中AOHP1为均一酸性杂多糖,AOHP2为精制多糖,由不同摩尔比的甘露糖(Man)、鼠李糖(Rha)、葡糖醛酸(GlcA)、半乳糖醛酸(GalA)、葡萄糖(Glc)、半乳糖(Gal)、木糖(Xyl)和阿拉伯糖(Ara)组成,量均相对分子量(Mw)分别为159.25和340.41,数均相对分子量(Mn)为131.378和42.861,多分散性指数(Mw/Mn)分别为1.21和7.94.实验表明在0~2 500μg·mL^(-1)浓度范围内两者对GES-1细胞无毒性作用,且呈现剂量依赖性的保护作用,其中AOHP2效果更优;当AOHP2≥250μg·mL^(-1)时,保护作用显著增强(P<0.05).研究证实AOHP具有胃保护作用,为高良姜资源的开发利用提供了科学依据和实验基础.
This study investigated the protective effect of Alpinia officinarum Hance Polysaccharides(AOHP)with different molecular weights and monosaccharide compositions against on GES-1 cell damage induced by ethanol.4 AOHP fractions(AOHP1~4)were obtained through decoction preparation and DEAE-52 anion-exchange chromatography.Chemical composition,molecular weight distribution,and monosaccharide composition were determined using phenol-sulfuric acid method,polarimetry,high-performance gel permeation chromatography(HPGPC),and high-performance liquid chromatography(HPLC).AOHP1 was identified as a homogeneous acidic heteropolysaccharide,while AOHP2 was a refined polysaccharide fraction composed of mannose(Man),rhamnose(Rha),glucuronic acid(GlcA),galacturonic acid(GalA),glucose(Glc),galactose(Gal),xylose(Xyl),and arabinose(Ara)in varying molar ratios.The weight-average relative molecular weights(Mw)were 159.25 and 340.41 kDa,number-average relative molecular weights(Mn)were 131.378 and 42.861 kDa,and polydispersity indices(Mw/Mn)were 1.21 and 7.94,respectively.MTT assays demonstrated that both fractions showed no cytotoxicity at concentrations of 0~2500μg·mL^(-1) and exhibited dose-dependent protective effects,with AOHP2 showing superior efficacy.Significant protective effects(P<0.05)were observed when AOHP2 concentration reached≥250μg·mL^(-1).The study confirmed that AOHP had the protective effect on the gastric mucosa,which provided the scientific basis and experimental foundation for the development and utilization of Alpinia officinarum Hance resources.
