硫酸化党参均一多糖 SCOP-2对肝癌HepG2细胞凋亡及BCL-2、BAX、p53基因表达的影响

摘要 目的 观察硫酸化党参均一多糖SCOP-2对肝癌细胞HepG2增殖、凋亡及凋亡相关基因的影响。方法 通过MTT法明确SCOP-2对HepG2细胞增殖的影响;采用Hoechst 33258染色荧光显微镜观察、流式细胞仪检查SCOP-2对HepG2细胞凋亡的影响;进一步通过RT-qPCR和Western blot检查SCOP-2对HepG2细胞凋亡相关指标BCL-2、BAX、p53及Caspase-3的影响。结果 实验结果表明,SCOP-2可以抑制HepG2细胞的增殖,促进细胞凋亡,其机制可能是SCOP-2作用后导致HepG2细胞BCL-2表达下调,BAX表达上调,进一步释放凋亡相关因子p53,再激活Caspase级联反应,促进Caspase-3表达和激活,从而促进HepG2细胞凋亡。结论 硫酸化党参均一多糖SCOP-2通过影响BCL-2、BAX、p53及Caspase-3的表达促进肝癌细胞的凋亡。 Objective To observe the effects of the sulfated homogeneous polysaccharide from codonopsis pilosula(SCOP-2)on the proliferation,apoptosis,and apoptosis-related genes of hepatocellular carcinoma HepG2 cells.Methods The MTT assay was used to determine the effect of SCOP-2 on the proliferation of HepG2 cells;Hoechst 33258 staining(observed under a fluorescence microscope)and flow cytometry were applied to detect the effect of SCOP-2 on the apoptosis of HepG2 cells;Reverse transcription-quantitative polymerase chain reaction(RT-qPCR)and Western blot were further used to examine the effect of SCOP-2 on apoptosis-related gene/proteins(BCL-2,BAX,p53,and Caspase-3)in HepG2 cells.Results The experimental results showed that SCOP-2 could inhibit the proliferation and promote the apoptosis of HepG2 cells.The possible mechanisms for SCOP-2 to act on HepG2 cells appeared to be due to the down-regulation of BCL-2 expression and the up-regulation of BAX expression in the cells,which further promoting the release of the apoptosis-related factor p53.Subsequently,the caspase cascade reaction is activated,which promotes the expression and activation of Caspase-3,thereby facilitating the apoptosis of HepG2 cells.Conclusion SCOP-2 promotes the apoptosis of HepG2 cells by regulating the expressions of BCL-2,BAX,p53,and Caspase-3.
