摘要
目的:探索新疆紫草根部可培养内生真菌的群落组成及在不同温度下分离培养获得内生真菌的多样性与不同产区真菌群落组成的差异性。方法:采用组织块法分离内生真菌,结合形态特征和分子生物学的方法进行综合鉴定,通过对比分析研究新疆紫草不同培养温度和不同地区内生真菌群落组成差异。结果:共获得516株内生真菌,划分为161个形态型,经ITS分子鉴定可划分为40个分类单元,归属2门5纲6目11科19属。子囊菌门(Ascomycota)为优势门,座囊菌纲(Dothideomycetes)为优势纲,格孢腔菌目(Pleosporales)为优势目,Chaetosphaeronema为优势属;15℃分离培养条件下获得的菌群丰富度、Shannon-Wiener指数(H′)、Simpson指数(D)、物种均匀度(E)均大于20℃和25℃分离培养条件下的对应值;巩留县、乌鲁木齐县南山、达坂城区、和静县分别获得25、11、11、4个分类单元。结论:新疆紫草根内生真菌具有丰富的物种多样性,分离温度对所获内生真菌群落组成、分离频率、丰富度以及多样性等都有影响,低温分离有利于获得更丰富的物种。不同产地的优势种群存在差异,巩留县的内生真菌群落组成最丰富。
Objective:To explore the community composition of culturable endophytic fungi from the roots of Arnebia euchroma,and the diversity of endophytic fungi obtained by isolation and culture at different temperatures and the differences in the composition of fungal communities in different production areas.Methods:Tissue block method was used to isolate endophytic fungi,combined with morphological characteristics and molecular biology for comprehensive identification,and comparative analysis was used to study the differences in the community composition of endophytic fungi at different isolation temperatures and in different regions of A.euchroma.Results:A total of 516 strains of endophytic fungi were obtained,divided into 161 morphotypes,which could be classified into 40 taxonomic units after ITS molecular identification,and were categorized into 2 phyla,5 classes,6 orders,11 families and 19 genera.Ascomycota was the dominant phylum,Dothideomycetes was the dominant class,Pleosporales was the dominant order,and Chaetosphaeronema was the dominant genus.The abundance of endophytic fungi,Shannon-Wiener index(H′),Simpson index(D),and species evenness(E)obtained at 15℃were greater than those at 20℃and 25℃;25,11,11,and 4 taxonomic units were obtained in Gongliu County,Nanshan of Urumqi County,Dabancheng District,and Hejing County,respectively.Conclusion:The endophytic fungi of A.euchroma had rich species diversity,and the isolation temperature affected the obtained endophytic fungal community composition,isolation frequency,richness,and diversity,etc.The low temperature isolation was favorable for obtaining richer species.The dominant populations differed among different origins,and Gongliu County had the richest endophytic fungal community composition.
