摘要
目的分析大连市不同来源副溶血性弧菌的毒力基因、耐药性等病原学特征和分子分型特征,为大连市食源性疾病风险监测预警和溯源提供数据支撑。方法收集2023年大连市腹泻患者粪便、食品及环境样本中分离出的54株副溶血性弧菌,经生化和飞行时间质谱鉴定,通过微量肉汤稀释法测定菌株的最低抑菌浓度(MIC)值,采用多重荧光聚合酶链反应检测tlh、tdh、trh基因,分型方法选用脉冲场凝胶电泳技术。结果54株副溶血性弧菌中,37株病例分离株有3株多重耐药菌,11株食品分离株也发现1株多重耐药株,6株环境分离株未发现多重耐药株。病例、食品、环境分离株tdh基因携带率分别为100.0%、63.6%和66.7%。聚类分析结果显示,大连市不同来源的副溶血性弧菌分散度较高(14.8%~100.0%),病例株带型相似度较高。结论2023年大连市检出的54株副溶血性弧菌分子分型分布呈现多态性,具有多重耐药株和食源性高毒力菌株,提示存在发生食源性疾病隐患。
Objective To analyze the pathogenic characteristics such as virulence genes and drug resistance,as well as the molecular typing features of Vibrio parahaemolyticus(VP)from different sources in Dalian City,providing data support for the risk monitoring,early warning and source tracing of foodborne diseases in Dalian.Methods A total of 54 VP strains isolated from fecal,food and environmental samples of diarrhea patients in Dalian City in 2023 were collected.These strains were identified by biochemical and time-of-flight mass spectrometry.The MIC values of the strains were determined by microbroth dilution method.The tlh,tdh and trh genes were detected by multiplex fluorescence polymerase chain reaction.Pulsed-field gel electrophoresis(PFGE)was used for typing.Results Among the 54 VP strains,no multi-drug resistant strains were found in the 6 environmental isolates,while 3 multi-drug resistant strains were identified among the 37 clinical isolates and 1 among the 11 food isolates.The carriage rates of the tdh gene in clinical,food and environmental isolates were all high,being 100.0%,63.6%and 66.7%respectively.The cluster analysis results showed that the VP strains from different sources in Dalian City were highly dispersed(14.8%-100.0%),and the banding patterns of clinical isolates were highly similar.Conclusion In 2023,the molecular typing distribution of 54 VP strains detected in Dalian City showed polymorphism,including multiple drug-resistant strains and highly virulent foodborne strains,suggesting the potential risk of foodborne diseases.
