NKA-Ⅱ型大孔树脂纯化菊花黄酮的研究

摘要 考察8种大孔树脂对菊花黄酮的静态吸附-解吸作用,筛选出纯化菊花黄酮的最佳大孔吸附树脂,以上样液质量浓度、上样液pH值、上样量、洗脱液浓度、洗脱液用量为影响因素,黄酮纯度为评价指标,进行工艺优化。结果表明,最佳工艺条件为NKA-Ⅱ型大孔树脂、上样液质量浓度为0.8~1.7 mg/mL、pH为4.24、上柱体积为2 BV、上样流量为1 mL/min,乙醇浓度为70%。用该最佳工艺进行验证试验,菊花黄酮的纯度由37.74%提升至64.28%。建立的工艺可为菊花在竹叶青酒及露酒方面的开发利用提供理论依据。 The static adsorption-desorption action of the total flavonoids of Flos Chrysanthemi by 8 kinds of macroporous resins were investigated to screen the best macroporous resin for purifying the flavonoids of Flos Chrysanthemi.The mass concentration,solution pH and upper column volume of the sample,the concentration of eluent and the elution volume as influence factors,and the purity of flavonoids as evaluation index were to optimize the process.The results indicated that the optimal process condition for separating the total flavonoids of Flos Chrysanthemi by NKA-II macroporous resin were as follows:the concentration of the sample was 0.8~1.7 mg/mL,pH of the sample solution was 4.24,the upper column volume was 2 BV,sample flow rate was 1 mL/min,the concentration of ethanol was 70%.The optimal process was used to verify the test,and the purity of the total flavonoids of Flos Chrysanthemi increased from 37.74%to an averaged of 64.28%.The established process can provide a theoretical reference for the development and application of Flos Chrysanthemi in Zhuyeqing Jiu and Lujiu.
