摘要
为探究不同溶剂对拐枣活性成分及生物活性的影响,采用蒸馏水、不同体积分数乙醇和6种低共熔溶剂(deep eutectic solvents, DESs)提取拐枣中活性成分,并对其抗氧化活性进行探究。结果表明:蒸馏水、体积分数60%乙醇、氯化胆碱-乙二醇(摩尔比1∶2、含水量30%,DES-3)的最佳料液比为1∶30、1∶40、1∶40(g/mL),其中DES-3在料液比为1∶40(g/mL)时提取的总黄酮、总多酚、多糖、生物碱和总皂苷含量,分别达(5.21±0.04)、(22.38±0.15)、(59.27±0.85)、(6.19±0.03)、(16.53±0.14) mg/g,高于乙醇和水。抗氧化能力分析结果表明,提取物中活性成分含量与抗氧化能力呈一定量效关系,DES-3提取物对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)自由基清除能力和总还原力优于体积分数60%乙醇和水。
In order to investigate the effects of different solvents on the active ingredients and antioxidant activities of Hovenia dulcis,distilled water,ethanol with different volume fractions and six deep eutectic solvents(DESs)were used to extract the active components of Hovenia dulcis,and their antioxidant activity was explored.The results showed that the optimal solid-liquid ratios for distilled water,60%ethanol by volume fraction,and DES-3 were 1∶30,1∶40 and 1∶40(g/mL),respectively.Among them,choline chloride-ethylene glycol(DES-3)with a molar ratio of 1∶2 and a water content of 30%showed the highest comprehensive content of active components when the solid-liquid ratio was 1∶40(g/mL).The contents of total flavonoids,polyphenols,polysaccharides,alkaloids,and saponins reached(5.21±0.04),(22.38±0.15),(59.27±0.85),(6.19±0.03)and(16.53±0.14)mg/g,respectively,which were higher than those extracted by ethanol and water.The result of antioxidant capacity revealed a certain dose-effect relationship between the content of active ingredients in the extracts and their antioxidant capacity.The DES-3 extract exhibited superior scavenging abilities against DPPH radicals and ABTS radicals,as well as total reducing power,compared to 60%ethanol by volume fraction and aqueous extracts.
