响应面法优化藏荆芥中E,Z-荆芥内酯冷浸提取工艺及活性评价

Optimization of Cold-soaked Extraction Technology of E,Z-nepetalactone from Nepeta Angustifolia C.Y.Wu by Response Surface Method and Evaluation of Its Activity

摘要 目的:采用响应面法优化乙醇冷浸提取藏荆芥中E,Z-荆芥内酯的工艺条件,并评价其降血压活性。方法:通过单因素考察,选择乙醇体积分数、液料比和提取时间为考察因素,E,Z-荆芥内酯转移率为响应值,采用Box-Behnken设计试验并进行响应面法分析,优化冷浸提取工艺。以卡托普利为阳性对照,通过血管紧张素转化酶抑制试验评价E,Z-荆芥内酯的体外降血压活性。结果:优化的最佳工艺为乙醇体积分数91%,液料比27∶1(mL∶g),提取时间47 h,在该最优条件下得到E,Z-荆芥内酯转移率为99.33%。活性评价结果表明E,Z-荆芥内酯对血管紧张素转化酶抑制率高达91.59%。结论:响应面法优化所得工艺科学合理、切实可行,可用于藏荆芥中E,Z-荆芥内酯的提取。E,Z-荆芥内酯具有较好的血管紧张素转化酶抑制活性,具有开发降血压药物的潜力。 Objective:To optimize the process conditions for cold soaking extraction of E,Z-nepetalactone from Nepeta angustifolia C.Y.Wu with ethanol using response surface methodology and evaluate its blood pressure lowering activity.Methods:The ethanol concentration,the liquid-material ratio and the extraction time were selected as examining factors by single factor experiment,and the transfer rate of E,Z-nepetalactone was selected as response value.The Box-Behnken design method and response surface methodology were used to optimize the cold-soaked extraction technology.Using captopril as a positive control,the angiotensinconverting enzyme inhibition test was performed to evaluate the in vitro blood pressure lowering activity of E,Z-nepetalactone.Results:The optimal process parameters for optimization are ethanol volume fraction of 91%,liquid to material ratio of 27∶1,and extraction time of 47 hours.The transfer rate of E,Z-nepetalactone with the optimal technology was 99.33%.The activity evaluation results indicate that E,Z-nepetalactone has a high inhibition rate of 91.59%on angiotensin-converting enzyme.Conclusion:The extraction technology optimized by response surface methodology was scientific,reasonable,and feasible,and it can be applicable for the extraction of E,Z-nepetalactone from Nepeta angustifolia C.Y.Wu.E,Z-nepetalactone has good angiotensin-converting enzyme inhibitory activity and has the potential to develop antihypertensive drugs.
