摘要
目的探讨诗碧曼精华液(SBM)对良性前列腺增生(BPH)的影响及其机制。方法将21只雄性6~8周龄ICR小鼠随机分为3组,每组7只。BPH组和SBM治疗组(SBM组)小鼠每天通过皮下注射丙酸睾酮进行造模,对照组(CON组)小鼠每天通过皮下注射等体积大豆油;从造模第2天开始每天于SBM组小鼠头部和背部涂抹SBM,于CON组和BPH组小鼠头部和背部涂抹等体积生理盐水,上述操作持续31 d后,分离各组小鼠血清和前列腺组织,以各组小鼠前列腺湿质量计算前列腺指数,通过苏木精-伊红(HE)染色观察各组小鼠前列腺组织形态学变化,通过ELISA法检测各组小鼠血清中双氢睾酮(DHT)、睾酮、雌二醇(E2)和前列腺酸性磷酸酶(PAP)的水平。向人前列腺增生上皮细胞系(BPH-1)中加入SBM进行处理,以加入等体积PBS处理的BPH-1作为对照,通过CCK-8法检测各组细胞增殖率,通过透射电镜及免疫荧光染色法观察BPH-1细胞内自噬小体数量,通过RT-qPCR和Western blot(WB)法检测各组细胞增殖、自噬和糖酵解相关分子的表达水平。结果与BPH组相比,SBM组小鼠前列腺指数显著降低,前列腺组织增生和炎性细胞浸润等病理改变均显著缓解,血清ACP、睾酮、DHT和PAP水平均显著降低,血清E2水平显著升高(F=4.96~1439.00,q=4.02~51.87,P<0.05)。与PBS处理的BPH-1细胞相比,SBM处理后BPH-1细胞的增殖率显著下降,Ki67 mRNA相对表达水平、PCNA mRNA和蛋白相对表达水平均显著降低,细胞内自噬小体数量显著增多,ATG7、ATG12、ATG16L1 mRNA相对表达水平以及ATG5、Beclin-1、HK2、PKM2、ALDOA mRNA和蛋白相对表达水平均显著升高(F=206.00~27234.00,q=16.99~325.50,t=8.73~82.28,P<0.05)。结论SBM可激活前列腺上皮细胞的糖酵解从而增强细胞自噬,进而缓解BPH。
Objective To investigate the effect of Shi-Bi-Man nourishing essence(SBM)on benign prostatic hyperplasia(BPH)and its mechanism.Methods A total of 21 male ICR mice aged 6-8 weeks were randomly divided into BPH group,SBM treatment group,and control group(CON group),with 7 mice in each group.The mice in the BPH group and the SBM treatment group were given subcutaneous injection of testosterone propionate every day to establish a model of BPH,and those in the CON group were given subcutaneous injection of an equal volume of soybean oil.Since day 2 of modeling,SBM was rubbed on the head and back of mice in the SBM group every day,and an equal volume of normal saline was rubbed on the head and back of mice in the CON group and the BPH group.After 31 consecutive days of treatment,serum and prostate tissue samples were collected,and the wet weight of prostate was measured to calculate prostate index;HE staining was used to observe histomorphological changes of the prostate;ELISA was used to measure the serum levels of dihydrotestosterone(DHT),testosterone,estradiol(E2),and prostatic acid phosphatase(PAP).The human BPH epithelial cell line(BPH-1)was treated with SBM,and the BPH-1 cells treated with an equal volume of phosphate-buffered saline(PBS)were established as the control group.CCK-8 assay was used to evaluate cell proliferation rate;transmission electron microscopy and immunofluorescent staining were used to measure the number of autophagosomes in BPH-1 cells;RT-qPCR and Western blot were used to measure the expression levels of molecules associated with cell proliferation,autophagy,and glycolysis.Results Compared with the BPH group,the SBM group had a significant reduction in prostate index,significant improvements in pathological changes such as prostatic hyperplasia and inflammatory infiltration,significant reductions in the serum levels of acid phosphatase,testosterone,DHT,and PAP,and a significant increase in the serum level of E2(F=4.96-1439.00,q=4.02-51.87,P<0.05).Compared with the BPH-1 cells treated with PBS,the BPH-1 cells treated with SBM had a significant reduction in proliferation rate,significant reductions in the relative mRNA expression level of Ki67 and the relative mRNA and protein expression levels of PCNA,and a significant increase in the number of autophagosomes,as well as significant increases in the relative mRNA expression levels of ATG7,ATG12,and ATG16L1 and the relative mRNA and protein expression levels of ATG5,Beclin-1,HK2,PKM2,and ALDOA(F=206.00-27234.00,q=16.99-325.50,t=8.73-82.28,P<0.05).Conclusion SBM can alleviate BPH by activating glycolysis and enhancing autophagy in prostate epithe-lial cells.
