负载聚多巴胺纳米粒子的多糖自愈合可注射水凝胶的生物相容性研究

摘要 动态自愈合可注射水凝胶作为一种新型的智能柔性生物材料,在组织工程和再生医学等领域展现出了广阔的应用前景.生物安全性研究对评价植入型水凝胶载体的实际应用价值至关重要.本研究全面评价了负载聚多巴胺纳米粒子的基于氧化透明质酸和N-羧乙基壳聚糖动态交联的自愈合可注射水凝胶的体外和体内生物安全性.体外生物相容性实验结果表明纳米复合动态水凝胶无细胞毒性且无体外溶血反应.将水凝胶注射至大鼠皮下检测了组织相容性和血液相容性,28天后观察主要脏器(心、肝、脾、肺、肾)的病理学切片表明无体内毒性,在大鼠尾动脉处取血,分析血常规、肝、肾功能等指标表明无血液毒性.这些研究结果为纳米复合动态水凝胶的进一步临床应用研究提供重要的理论和实验基础. Dynamic self-healing injectable hydrogel emerged as a new intelligent flexible biomaterial,showing broad applications in tissue engineering,regenerative medicine and other fields.Biosafety is very important to evaluate the practical application value of implantable hydrogels.This study comprehensively evaluated the in vitro and in vivo biological safety of a self-healing injectable hydrogel based on hyaluronic acid and N-carboxyethyl chitosan loading with polydopamine nanoparticles.The results of in vitro biocompatibility test showed that the nanocomposite dynamic hydrogel had no cytotoxicity and hemolysis in vitro.The histocompatibility and blood compatibility were tested by injecting the hydrogel under the skin of rats.After 28 days of observation,the pathological sections of the main organs(heart,liver,spleen,lung,kidney)demonstrated no toxicity in vivo.Blood was taken from the tail artery of rats to analyze blood routine,liver and kidney functions showed no toxicity in vivo.These results provide an important theoretical and experimental basis for further clinical applications of nanocomposite dynamic hydrogels.
