摘要
目的检测胰腺癌组织中焦亡相关基因焦孔素C(GSDMC)、焦孔素D(GSDMD)和NLR家族含CARD结构域4(NLRC4)表达水平,并探究其与患者临床病理特征和预后的关系。方法选择2019年12月至2022年3月在南京医科大学第四附属医院接受胰腺癌根治术治疗的86例胰腺癌患者作为研究对象,收集其经手术切除的胰腺癌组织和癌旁组织(距离肿瘤边缘>2 cm)标本。分别采用实时荧光定量PCR法、免疫组织化学染色法检测组织中GSDMC、GSDMD和NLRC4的mRNA和蛋白表达水平,并分析其与患者临床病理特征的关系。采用KaplanMeier生存曲线分析胰腺癌组织中GSDMC、GSDMD和NLRC4蛋白表达与患者预后的关系。结果与癌旁组织相比,胰腺癌组织中GSDMC、GSDMD和NLRC4的m RNA表达水平均显著升高,差异均有统计学意义(P均<0.05)。与肿瘤最大径>2 cm、中~高分化、TNM分期为Ⅰ期的胰腺癌患者相比,肿瘤最大径>2 cm、低~未分化、TNM分期为Ⅱ期的患者的胰腺癌组织中GSDMC和NLRC4的m RNA表达水平均显著升高(P均<0.05);与中~高分化、TNM分期为Ⅰ期的胰腺癌患者相比,低~未分化、TNM分期为Ⅱ期的患者的胰腺癌组织中GSDMD m RNA表达水平均显著升高(P均<0.05)。Kaplan-Meier生存曲线分析结果显示,GSDMC蛋白阳性组的3年总生存率为17.78%(8/45),显著低于GSDMC蛋白阴性组(39.02%,16/41),差异有统计学意义(P<0.05)。结论GSDMC、GSDMD和NLRC4在胰腺癌组织中均表达上调,且其中GSDMC蛋白阳性表达与胰腺癌患者预后不良密切相关。
Objective This paper aims to detect the expression levels of pyroptosis-related genes—Gasdermin C(GSDMC),Gasdermin D(GSDMD),and NLR family CARD domain containing 4(NLRC4)in pancreatic cancer tissue,and to analyze their relationship with clinicopathological characteristics and prognosis.Methods A total of 86 patients who underwent radical resection of pancreatic cancer in the Fourth Affiliated Hospital of Nanjing Medical University from December 2019 to March 2022 enrolled in the study.Pancreatic cancer tissues and adjacent non-tumorous tissues(>2 cm from the tumor margin)removed during surgery were collected.The mRNA and protein expression levels of GSDMC,GSDMD and NLRC4 in the tissues were detected by real-time fluorescence quantitative PCR and immunistochemical staining,respectively,and their relationship with the clinicopathological characteristics of patients was analyzed.Kaplan-Meier survival curves were used to analyze the relationship between protein expression of GSDMC,GSDMD,and NLRC4 in pancreatic cancer tissues and patient prognosis.Results Compared with adjacent non-tumorous tissues,the mRNA expression levels of GSDMC,GSDMD,and NLRC4 in pancreatic cancer tissues are significantly higher,with significant statistical differences(P<0.05).Compared with patients with a maximum tumor diameter≤2 cm,moderately to highly differentiated tumors,and TNM stageⅠtumors,the mRNA expression levels of both GSDMC and NLRC4 in the pancreatic cancer tissues of patients with a maximum tumor diameter>2 cm,poorly to undifferentiated tumors,and TNM stageⅡtumors are significantly increased(P<0.05).Compared to patients with moderately to highly differentiated tumors and TNM stageⅠtumors,the mRNA expression levels of GSDMD in the pancreatic cancer tissues of patients with poorly to undifferentiated tumors and TNM stageⅡtumors are significantly increased(P<0.05).Kaplan-Meier survival analysis shows that the 3-year overall survival rate is 17.78%(8/45)in the GSDMC protein-positive group,which is significantly lower than that in the GSDMC protein-negative group(39.02%,16/41),with a significant statistical difference(P<0.05).Conclusion GSDMC,GSDMD,and NLRC4 are all up-regulated in pancreatic cancer tissue,and positive expression of GSDMC protein is closely related to poor prognosis in patients with pancreatic cancer.
