摘要
目的 通过薄层色谱鉴别和同条件下3种活性成分的含量测定建立活血跌打酒的质量标准。方法 以薄层色谱法(TLC)建立大黄、黄柏、两面针、当归尾及桂枝的定性鉴别方法。同时利用高效液相色谱法(HPLC)对阿魏酸、盐酸小檗碱及氯化两面针碱3种成分进行含量测定。结果 大黄、黄柏、两面针、当归尾及桂枝的薄层色谱斑点分离效果较好,斑点显色清晰且无干扰,具有良好的特征性。阿魏酸、盐酸小檗碱、氯化两面针碱分别在3.92~39.2μg/mL,35.99~359.9μg/mL和2.47~24.7μg/mL范围内具有良好的线性关系;平均回收率分别为103.85%、98.39%和103.36%。结论 本研究建立的薄层色谱鉴别方法和含量测定方法简单、可行、专属性强,适用于活血跌打酒的质量控制,研究结果可为后续研究与开发提供支持。
Objective To establish the quality standard for Huoxue Dieda Medicinal Liquor by identification via thin-layer chromatography and determination of the contents of three active ingredients under the same conditions.Methods TLC was used to establish the qualitative identification methods for Rheum officinale Baill.(Dahuang),Phellodendron chinense Schneid.(Huangbo),Zanthoxylum nitidum(Roxb.)DC.(Liangmianzhen),Angelica sinensis(Oliv.)Diels root tail(Dangguiwei),and Cinnamomum cassia Presl twig(Guizhi).Meanwhile,HPLC was used to determine the contents of ferulic acid,berberine hydrochloride and nitidine chloride.Results The TLC spots of Rheum officinale Baill.,Phellodendron chinense Schneid.,Zanthoxylum nitidum(Roxb.)DC.,Angelica sinensis(Oliv.)Diels root tail,and Cinnamomum cassia Presl twig.were well separated,with clear coloration and no interference,showing good specificity.Ferulic acid,berberine hydrochloride,and nitidine chloride showed good linear relationships in the concentration ranges of 3.92-39.2μg/mL,35.99-359.9μg/mL,and 2.47-24.7μg/mL respectively;the average recovery rates were 103.85%,98.39%and 103.36%respectively.Conclusion The TLC identification method and content determination method established in this investigation are simple,feasible,and highly specific,which are suitable for the quality control of Huoxue Dieda Medicinal Liquor.The research results can provide support for subsequent research and development.
