PLAC8激活ERK信号通路促进肺腺癌PC-9细胞增殖

Cell proliferation of lung adenocarcinoma PC-9 promoted by PLAC8 through activation of Erk signaling pathway

摘要 目的探讨胎盘特异性蛋白8(PLAC8)对肺腺癌细胞增殖的影响。方法构建PLAC8干扰及PLAC8过表达的肺腺癌(LUAD)细胞株PC-9,Western blot方法验证PLAC8干扰及过表达效果;CCK-8方法检测PC-9细胞增殖;Western blot方法检测PC-9细胞ERK信号通路蛋白表达;在PLAC8过表达、PLAC8干扰及相应对照组PC-9细胞中加用ERK信号通路抑制剂U0126,CCK-8方法检测各组细胞增殖能力变化。结果干扰PLAC8的表达导致肺腺癌细胞增殖能力降低,而过表达PLAC8则提高肺腺癌细胞的增殖;抑制ERK信号可显著降低过表达PLAC8导致的细胞增殖能力增强。结论PLAC8能通过激活ERK信号通路促进肺腺癌细胞PC-9的增殖。 Objective To investigate the effect of placenta specific protein 8(PLAC8)on the proliferation of lung adenocarcinoma cells..Methods Plac8-interfered and PLAC8-overexpressed lung adenocarcinoma(LUAD)cell line PC-9 was constructed,and the effects of PLAC8-interfered and overexpressed PLAC8 were verified by Western blot.PC-9 cell proliferation was detected by CCK-8 method.The expression of ERK signaling pathway protein in PC-9 cells was detected by Western blot.PLAC8 overexpression,PLAC8 interference and corresponding control group PC-9 cells were treated with ERK signaling pathway inhibitor U0126,and CCK-8 method was used to detect the changes of cell proliferation ability in each group.Results Knockdown of PLAC8 inhibited decreased the proliferation of lung adenocarcinoma cells,while overexpression of PLAC8 increased the proliferation of lung adenocarcinoma cells.Inhibition of ERK signaling significantly reduced cell proliferation induced by overexpression of PLAC8.Conclusion PLAC8 could promote the proliferation of PC-9 cells by activating ERK signal pathway.
