摘要
目的探究脂肪干细胞(ADSCs)对糖尿病足溃疡大鼠创面愈合的影响及机制。方法将40只糖尿病足溃疡Wistar大鼠随机分为模型组、ADSCs组、ADSCs+antagomiR-NC组和ADSCs+antagomiR-23a-3p组,每组10只,计算大鼠创面愈合率,RT-qPCR检测大鼠创面组织中miR-23a-3p和JAZF1 mRNA表达;Masson染色检测大鼠创面组织中胶原纤维含量;免疫荧光检测大鼠创面组织中VEGFA表达;Western blot检测大鼠创面组织中JAZF1蛋白表达,采用双荧光素酶报告基因实验验证miR-23a-3p与JAZF1靶向关系。结果ADSCs移植增加DFUs大鼠创面愈合率以及创面组织中胶原沉积和VEGFA表达,降低大鼠创面组织中JAZF1表达;抑制miR-23a-3p部分逆转ADSCs移植对DFUs大鼠创面愈合和胶原沉积的促进作用以及VEGFA表达的上调作用,并部分逆转ADSCs移植对DFUs大鼠创面组织中JAZF1表达的下调作用。结论ADSCs移植通过分泌miR-23a-3p促进DFUs大鼠创面愈合,其机制可能与靶向抑制JAZF1表达有关。
Objective To explore the effect and mechanism of adipose derived stem cells(ADSCs)on the wound healing of diabetic foot ulcer rats.Methods Forty Wistar rats with diabetic foot ulcer were randomly divided into model group,ADSCs group,ADSCs+antagomiR-NC group and ADSCs+antagomiR-23a-3p group,with 10 rats in each group.The wound healing rate of rats was calculated,and the mRNA expressions of miR-23a-3p and Jazfl were detected by RT qPCR.The content of collagen fibers in rat wound tissue was detected by Masson staining.The expression of VEGFA in the wound tissue of rats was detected by immunofluorescence.The expression of JAZF1 protein in the wound tissue of rats was detected by Western blot.The targeting relationship between miR-23a-3p and JAZF1 was verified by double luciferase reporter gene experiment.Results ADSCs transplantation increased the wound healing rate,collagen deposition and the expression of VEGFA in the wound tissue of DFUs rats,and decreased the expression of JAZF1 in the wound tissue of rats.Inhibition of miR-23a-3p partially reversed the promotion of ADSCs transplantation on wound healing and collagen deposition and the up regulation of VEGFA expression in DFUs rats,and partially reversed the down regulation of JAZF1 expression in wound tissue of DFUs rats by ADSCs transplantation.Conclusion ADSCs transplantation promotes wound healing in DFUs rats by secreting miR-23a-3p,and its mechanism is related to the targeted inhibition of JAZF1 expression.
