娜塔栎种子萌发过程中营养成分及内源激素含量变化与转录代谢分析

Analysis of the Changes in Nutrient Components and Endogenous Hormone Contents and Transcriptional Metabolism During the Germination Process of Quercus texana Seeds

摘要 以娜塔栎种子为试验材料,测定种子不同萌发阶段的淀粉、还原糖、可溶性糖和可溶性蛋白以及内源激素(ABA、IAA、MeIAA、JA、SA)含量变化,并结合转录组学数据进行分析。结果表明,娜塔栎种子萌发属子叶留土型,萌发过程分为吸胀、萌动、发芽和幼苗等4个阶段。营养物质呈动态变化:蛋白先降后升,淀粉持续下降,可溶性糖和还原糖先升后降再升。激素脱落酸(ABA)、吲哚乙酸(IAA)、IAA甲酯(MeIAA)含量先降后升,茉莉酸(JA)含量先升后降再升,水杨酸(SA)含量先降后升,萌动阶段为关键转折期。相关性分析显示,发育阶段与营养物质负相关,蛋白、淀粉和可溶性糖与激素间显著负相关,还原糖与激素正相关,激素间正相关。主成分分析确定SA和还原糖为关键指标。对娜塔栎发育过程进行转录代谢分析,结果表明,差异基因主要富集在碳水化合物、细胞多糖及糖基转移酶中,表明娜塔栎种子萌发所需大量能量来源于淀粉、还原性糖等;联合激素代谢分析结果,JA、Auxin类激素在娜塔栎种子萌发过程中表现活跃,为娜塔栎种子萌发提供能量。种子营养物质及转录代谢组水平揭示了娜塔栎种子的萌发机制。 Using the seeds of Quercus texana as the experimental materials,the changes in the contents of starch,reducing sugar,soluble sugar,soluble protein and endogenous hormones(ABA,IAA,MeIAA,JA,SA)at different germination stages of the seeds were determined,and analyzed in combination with transcriptomic data.The results showed that the seed germination of Quercus texana belonged to the cotyledon soil-retaining type,and the germination process was divided into 4 stages:swelling,awakening,germination and seedling.Nutrients showed dynamic changes:protein first decreased and then increased,starch continued to decline,and soluble sugar and reducing sugar first increased,then decreased,and then increased again.The contents of hormones abscisic acid(ABA),indoleacetic acid(IAA),and methyl IAA(MeIAA)first decreased and then increased,JA first increased,then decreased,and then increased again,SA first decreased and then increased.The budding stage was a critical turning point.Correlation analysis showed that the developmental stage was negatively correlated with nutrients,while protein,starch and soluble sugar were significantly negatively correlated with hormones.Reducing sugar was positively correlated with hormones,and there was a positive correlation among hormones.Principal component analysis identified SA and reducing sugars as key indicators.Transcriptional metabolic analysis of the development process of Quercus texana showed that the differential genes were mainly enriched in carbohydrates,cellular polysaccharides and glycosyltransferases,indicating that the large amount of energy required for the germination of Quercus texana seeds came from starch,reducing sugars,etc.The results of the combined hormone metabolism analysis showed that JA and Auxin hormones were active during the germination process of Quercus texana seeds,providing energy for their germination.The levels of seed nutrients and transcriptional metabolomics revealled the germination mechanism of Quercus texana seeds.
