摘要
对7-氨基去乙酰氧基头孢烷酸亚砜(简称7-ADCA亚砜)的合成工艺进行优化改进。以去乙酰氧基头孢菌素C(简称DAOC)为原料,经氧化、酶催化水解、结晶等途径得到7-ADCA亚砜。利用1H-NMR和MS对产物进行结构鉴定。优化后的工艺成效显著,总收率提升至80%以上,纯度经HPLC法测定达99.70%。新路线避免使用有毒有害的物料与试剂,反应条件更为温和环保,且杂质更加可控,操作更为简便,适合工业化生产。
The synthesis process of 7-aminodeacetoxycephalosporanic acid sulfoxide(7-ADCA sulfoxide)was optimized and improved.7-ADCA sulfoxide was synthesized from deacetoxycephalosporin C(DAOC)by oxidation,enzymatic hydrolysis and crystallization.The structure of the product was identified by 1H-NMR and MS.The optimized process achieved significant results,with a total yield of over 80%and a purity of 99.70%as determined by HPLC.The new route avoided the use of toxic and harmful materials and reagents,with milder and more environmentally friendly reaction conditions,more controllable impurities,and easier operation,making it suitable for industrial production.
